With the change in the rabbit industry produced increasing levels of consumer demand and the market, meat quality traits becoming breeding research focus in future. Meat quality can be influenced by genetic, nutritional levels and living environment and other factors, the molecular mechanisms of environmental regulation on muscle growth traits in the epigenetic level, to improve the quality of animal meat and meet the demand for meat safety has positive significance. Myo G gene Myo D gene family is the only gene in all cell lines can be expressed in skeletal muscle, is required for skeletal muscle differentiation factor, which to play a key role in muscle differentiation by controlling the muscle cells to form fusion and muscle fibers. Therefore, this study was designed to take advantage of Myo G gene as a candidate gene epigenetic molecules present study, the use of BSP and Q-PCR as the main molecule experiments Methods.Qingdao Kangda Rabbit Development Co., Ltd. and Shandong Agricultural University jointly cultivate 84 days rabbit age groups supporting system for the test, the Conventionenal environment in which rabbits and SPF environment rabbits and female, as the test material to study the effects of different levels of environmental performance and Myo G rabbit gene expression and methylation frequency and slaughter correlation analysis traits. The main results are as follows:(1)Two different environments Zealand rabbit slaughtering production traits analysis showed that SPF environment rabbits body weight, eviscerated weight, legs weight was significantly higher than in Conventionenal environment of New Zealand rabbits(P < 0.05);SPF New zealand rabbits major organs compared with the normal level rabbits, although no significant difference between the heart and lungs(P > 0.05), the rest of the several organs of SPF were significantly lower than the Conventionenal level of New Zealand rabbits(P <0.05).(2)For two different grades environments rabbits longissimus muscle slice experiments,analysis of differences between the two environments rabbits muscle fiber traits of SPF rabbits showed that muscle fiber diameter is significantly smaller than an Conventionenal environment(P < 0.05), the density is significantly greater than the Conventionenalenvironment(P < 0.05), presumably of SPF rabbits muscles than ordinary environment rabbits tender, juicy and delicious.(3)New Zealand rabbits two different grades environment hind legs, longissimus fluorescence quantitative analysis showed that the expression level of gene expression hind Myo G SPF environment of New Zealand rabbit was significantly higher than the Conventionenal environment of the hind legs(P < 0.05), SPF environment New Zealand rabbit Beiyao Myo G gene expression was significantly lower than the Conventionenal environment(P < 0.05), but also found that the expression of both hind legs in both environments > Beiyao expression levels laws.(4)The two environments and rabbits hind legs are dorsi muscle were Myo G gene promoter region upstream of the transcription start site that is to methylation analysis 595 bp downstream region of about 500 bp, find transcripts as a whole upstream of the start point-80 bp to + 506 bp downstream region in a high methylation state( > 60%); promoter-331 bp to-61 bp region in hypomethylation(< 40%); Myo G gene promoter region-593bp-346 bp to moderate methylation status in the region(40%-60%). And found two environments rabbits hind legs in Myo G gene promoter region-80 bp to + there are significant differences in the frequency of methylation 328 bp, that Myo G gene promoter-80 bp to + 328 bp area, SPF environment after rabbit methylation rate leg is significantly less than the Conventionenal environment rabbit hind methylation ratio(P < 0.05); two environments rabbits Beiyao in Myo G gene promoter region methylation frequency-80 bp to + 506 bp of the obvious differences that Myo G gene promoter-80 bp to + 506 bp area, SPF environment rabbit Beiyao methylation was significantly higher than the Conventionenal environment rabbit Beiyao methylation ratio(P < 0.05).(5)By Myo G gene transcription factor promoter region and its associated binding site prediction can then analyze their gene expression in Moy G methylation through relevant research and analysis, suggesting that AP-2, NF1 F, E2 F transcription factors, etc. relations plays a role can not be ignored, SP1, CTCF, YY1 F transcription factors belonging to DNA methylation-dependent non-binding factor may be the relationship between gene methylation and its expression Moy G effect is not great. By this analysis, the better to further illustrate the interaction between DNA methylation and its corresponding mechanisms between the expression amounts.These results can be assumed that levels of cleanliness in the environment to a certain extent can affect meat rabbit production traits through DNA methylation epigenetic modification ofmolecular mechanisms, and speculated that the methylation status of the model for future gene may Myo G molecular markers molecular breeding.
