Coding sequence(CDS) and 3'untranslated sequence of Myogenin(MyoG) gene from rabbit was cloned and sequenced for the first time(GenBank Accession Number: FJ548758,FJ605115,FJ605116,FJ608376).The structure and function of MyoG were predicted by bioinformatics methods;Single nucleotide polymorphism in the coding sequence of Myogenin(MyoG) gene was detected by PCR-SSCP and DNA sequencing in 6 breeds including Haerbing white rabbit,hoto rabbit,Tianfu black rabbit,Belgian hare, ZIKA rabbit and California rabbit,and the effect of genotypes on performance traits was studied by GLM analysis.The results showed:Fragment length of Myogenin(MyoG) gene was 2331 bp by cloning and sequencing, contents exon1(partial),intron1,exon2,intron2 and exon3(partial).It is showed that MyoG gene have high homology among different species including swine(NM-001016725),human(NM-002479),macaque(XM-001104569),Mus(NM-031189),Rattus(NM-017115),cattle(001111325),canis familiaris(XM-547345),chicken(NM-204184),zebra fish(NM-131006),Bufo(NM-001016725) by Multiple Sequences Alignment(MSA),and the highest nucleotide homology with rabbit is swine and canis familiaris.The structure and function of MyoG were predicted by bioinformatics methods,seven potential N-glycosylation sites in MyoG amino acid sequence was found,site98,111,113,140,176,183,198 could be glycosylated.Four main hydrophobicity region of MyoG were found,which stand 65-70,120-126,132-138 and 202-219 in amino acid sequence respectively.Fourteen potential phosphorylation sites in MyoG amino acid sequence were found:Ser43,Ser77,Ser79,Ser165,Ser167,Ser171,Ser204,Ser208,Ser215,Thr87,Thr152,Tyr4,Tyr22 and Tyr35.Three disulfide bond of MyoG formed between Cys61 and Cys73,Cys65 and Cys70,Cys163 and Cys170 respectively,in order to maintain spatial structure of MyoG.The three dimensional structure of MyoG based on fourα-helices.Fourα-helices formed by 67 amino acid residues with amount of 29.78%of MyoG sequence; the interval amongα-helices standed by random coil which formed by 158 amino acid residues with amount of 70.22%of MyoG sequence.2 SNPs including exon1+356and exon3+232was studied by PCR-SSCP in 6 rabbit.It is showed that 2 allele(denoted as A and B) and 3 genotype(denoted asAA,AB and BB)was detected in 356 bp site of exon1,the frequency of 2 allele and 3 genotype was 0.632,0.368,0.275,0.560,0.165 respectively,PIC value of six group was higher than 0.25, is medium polymor.2 allele(denoted asM and N) and 3 genotype(denoted as MM, MNand NN)was detected in 232bp site of exon3,the frequency of 2 allele and 3 genotype was 0.544,0.456,0.310,0.510,0.180,PIC value of six group was higher than 0.25,is medium polymor,is medium polymor.In the analysis of the correlation between MyoG gene and the growth trait by using the data of these six breeds respectively,at locus exon1+356,the least squares means of Body weight,Visceraste weight and Visceraste weight percentage had significant difference between AA and BB genotype((P<0.05),half-Visceraste weight percentage and Cooked-meat ratio have no significant difference with three genotype(P>0.05);at locus exon3+232,the least squares means of Visceraste weight and Visceraste weight percentage had significant difference between MM and MN genotype((P<0.05),the least squares means of Body weight,Half-Visceraste weight percentage and Cooked-meat ratio have no significant difference between MM,MN and MN genotype(P>0.05);MyoG gene could be a candidate modifier gene that affect or control the body weight and production traits of rabbits.
