Study On Technology Optimisation Of Sexual Reproduction And Tissue-Culture Shoots Transplant Of Dendrobium Officinale

Dendrobium officinale was one epiphytes orchids, belong to Dendrobium, was the rare traditional Chinese medicin contained into the "Chinese pharmacopoeia" of 2010 edition. Dendrobium officinale had significant effect on anti-aging, enhanceing immune function, the anti tumor, diabetes, cataract and rehabilitation treatment of weak patients. As the depth development of Dendrobium officinale products, wild resources were dried up almostly, however, the market gap was very big, artificial planting scaley developed. Guangxi was one of the producing of Dendrobium officinale, and also was an emerging region in China. At present, under the condition of in vitro, seeds would be an effective way to expanding Dendrobium officinale seedlings number. This study on technology optimisation of sexual reproduction and tissue-culture shoots transplant of Dendrobium officinale, was to obtain a set of artificial breeding method, and optimization of breeding strong seedlings and artificial transplanting technology, in order to providing certain theoretical basis and technical support for producers. The results showed that:(1) The 3-4 d after flowering was good pollination times of Dendrobium officinale, abortive generally appeared in the 3-4 d after pollination, the fruit growed fastest within 20 d after artificial pollination, and the 120-150 d was mature period.The best medium for Dendrobium officinale seeds germination was Hyponex NO.1 3 g/L+sugar20 g/L+agar4.8g/L+coconut juice 100ml/L.(2) 1/2 MS had significant effect on seedling when was the basic culture medium, but Hyponex, MS were poorer. Bananas were good for seedlings growth,60 g/Lbananas effected on seedlings much; Coconut water combination effect was relatively poor.Lv feng yuan could effectively promote develop of Dendrobium officinale somaclone growth. The good seedling culture medium was 1/2MS+sugar 20 g/L+banana 60 g/L+ peptone 1 g/L+agar 5g/L+active carbonlg/L+Lv fengyuan 10 ml/L. Keep roots was good for the process of biomass accumulation, but cutting roots was easier to form new roots.(3) Between April to June, and September to October, which two season were best time of transplanting seedlings for the Dendrobium officinale in guangxi.Open smelting could made appropriate seedling water loss,1-2 d was adapt to the environment gradually, was beneficial to the growth of the somaclone after transplanting.Strong seedlings were good for transplanting survival of Dendrobium officinale, and plant height, root number, root length were important factors. With 3 roots, root length 5 cm, plant height 5 cm above, which the somaclone plant grew much better, had advantage for nutrient reserves of seedlings and roots activity.Transplanting substrate should be based on the principles of air, water, and mixed matrix could be better for plant growth, adding peat substrate was best for the grow of Dendrobium officinale, the worst effect substrate was the mixed matrix adding wood chaff matrix, which not only the survival rate was low, but plant growth was poor.
