Salmonella is enterobacteriaceae, serotype is various, it does a lot of damage to human and livestock. In recent years, poultry farmers and public health had pay attention to the problem of salmonella contaminated livestock products. With the use or misuse of antibiotics, making the drug resistance of salmonella more and more serious, pose a great threat to the breeding.This experiment successively from the different cities of shandong province to collect different samples of animal origin, by selective media, gram staining, biochemical test and 16 s r RNA to determine whether the separation of salmonella. Through the slide agglutination test, determine the serotype. By mircrobroth dilution assay to detect the drug resistance of the isolated strain. While detecting the 17 resistance gene of the isolated strain. Determination of four common virulence genes of salmonella, understand the characteristics of the separation of salmonella virulence gene mutation. Through the MLST, understand the gene sequence type of salmonella.We abtain a total of 78 chicken salmonella strains, 56 duck salmonella strains and 20 pigs salmonella strains.The result of different animal origin Salmonella serotype: chicken origin have 6 kinds of serotype, salmonella enteritidis accounted for 88.5%; Salmonella typhimurium accounted for 1.3%; Salmonella Senftenberg accounted for 1.3%; salmonella Indiana accounted for 5.1%; Salmonella thompson accounted for 2.6%; Salmonella agama accounted for 1.3%. Duck origin have two serotype, salmonella enteritidis accounted for 67.9%; Salmonella typhimurium accounted for 32.1%. Pig origin have three kinds of serotype, salmonella typhimurium most, accounting for 65%; Salmonella enteritidis accounted for 15%; Salmonella derby accounted for 20%. Through MLST technology, 7 kinds of ST type: chicken origin detection: ST11, ST19, ST26, ST128, ST14, ST17 and New1, duck origin have three type: ST ST11, ST19 and New2, pig origin have three type: ST34, ST40 and ST3007.Through the drug sensitive test, in the chicken origin strains, ampicillin and polymyxin, are the highest drug resistance rate, reached more than 80%, the ciprofloxacin and cefuroxime drug resistance rate reached 59.0% and 57.7%. Duck origin strains, the ciprofloxacin and polymyxin B resistance rate reached 100%, the enrofloxacin resistance rate reached 83.9%, doxycycline, gentamicin and neomycin resistance is higher than 60%. The swine origin strains, the doxycycline resistance rate reached 95%, followed by ampicillin was 80% and florfenicol was 45%. In the statistical analysis found that significant difference phenomenon of the resistance rate between the different animal origin is widespread. In the resistance gene detection, qnr A and qnr B genes were higher than 70% in three kinds of animal origin, the second is tet A, in the duck origin was 42.9%, in the other two kinds of animal origin were higher than 50%, the drug resistance gene bla TEM detection rate were higher than 30% in the three kinds of animal origin, moreover bla SHV, aph(3 ')- IV, tet B, qnr C and qep A genes detection rate to zero in three kinds of animal origin. In the statistical analysis found that significant difference phenomenon of the resistance gene rate between the different animal origin is nfrequent.Virulence gene detects found four virulence genes homologous rates are high, but there some base mutation, the salmonella of swine origin has not been mutated. inv A gene: chicken and duck has two main locus mutation, but did not change in amino acid levels. hil A gene: only chicken origin in two sites mutation, amino acid comparison, found that in the base 199- bit mutation lead to amino acid change. sii E gene: only chicken origin in four sites mutation, the mutations in open reading frame, did not happen the variation of amino acid level. inv H found no base mutation.This experiment analysis the characteristics of the different animal origin salmonella from the multiple level, which enables us to have a preliminary understanding about the different animal origin salmonella in shandong province, and is helpful to better prevention and treatment of the infections caused by bacteria, and targeted to make preventive measures.
