Iodine has a powerful effect on killing bacteria, also can kill bacterial spore, fungus, virus, protozoon, and so on. Iodine is mainly dependent on the form of molecules(I2) to realize its function of killing bacteria, with its principle is to iodide or oxidize the active gene of bacteria protein and combine with protein amino to make protein degenerative and inhibit the bacteria metabolic enzyme system. Many disinfectants that Iodine was made as the main ingredient has appeared since the Iodine was discovered. Due to its low water solubility and its volatility make it greatly limits on its application range. However, ?-cyclodextrin and its derivatives' unique cavity structure which could pack in a variety of drugs. After packing with the ?-cyclodextrin, the water solubility, the use ratio and the stability of iodine are obviously increased. On the other hand, the ?-cyclodextrin is an environmental compound which is green, natural and biodegradable.At present, the breeding industry in our country is stepping towards scale and intensivism. The large density and the terrible environment make it more possible for the occurrence of diseases, and many of the diseases are caused by the existence of pathogenic microorganisms in the environment. In order to control the spread of disease, cut off thesources of infection and reduce the economic loss caused by the large scale epidemic, as we all know, disinfection is the essential process. The purpose of this test is finding out a kind of disinfectant that can enhance the Iodine's stability, have good disinfection effect, as well as decrease the pollution and cost, and be suitable for the disinfection in animal buildings.In this test, the ?-cyclodextrin complexation iodine was used to improve the water solubility, the stability and the disinfection effect. After analysing the manufacturing technique of the ?-cyclodextrin complexation iodine, the best process route was determined. The quality analysis of samples including the character, identification, examination and content determination, and the test of antiviral and antibiosis were inspected at the same time. And the study of the condition of different temperature, pH and organic matter. About twenty kinds of organic solvents were compared to choose the best solvents for Iodine. Finally, the anhydrous ethanol was regard as the best solvent for iodine. Through repeating preliminary tests and the reference literature, according to the preparation method of ?-cyclodextrin complexing with Iodine, four influential factors were chosen which were the molar ratio of ?-cyclodextrin and Iodine, stirring speed, stirring time and temperature of ?-cyclodextrin saturated solution mixing with Iodine I as our target. With three levels for every factor, test method with L9(34) orthogonal test table was designed, and the examination indicators with effective Iodine was made, Iodine yield, comprehensive weighted mean determined, and got the optimal techological route in the end.The best testal data received through the method of L9(34) orthogonal test table were that the stiring speed was 500 r/min, the molar ratio of ?-cyclodextrin and Iodine was 1:1, stiring time was 8 h, temperature of ?-cyclodextrin saturated solution mixing with Iodine was 75?. We use the method of sodium thiosulfate oxidation-reduction to detect the content of the ?-cyclodextrin complexation iodine. The linear equation of the concentration of iodine and the precision was 0.08% ± 0.01% and the accuracy was 93.86%- 98.08%. The content of effective iodine was above 16% on average. the item of dry weight loss also was checked, determination of iodine ions, residue on ignition and heavy metal inspection. The dry weight loss received from the test should not be greater than 3%, and in the checking of residue on ignition we received that the color of a pipe was shallow than the color of a pipe, and all content of heavy metal was not greater than 10%. What's more, we also found that the ?-cyclodextrin complexation iodine did well in killing escherichia coli, bacillus, staphylococcus aureus, NDV and IBDV.This test finally proved that the ?-cyclodextrin complexation iodine is successfully. With its advantages of simpleness in process, good quality in control, stability, better microbial killing effect, and the ?-cyclodextrin will have a good development prospect.
