| In this study, Yili horses were used for the main experiment object.DMRT3,MSTN and ACTN3 gene were chosen as main candidate genes.Genetic polymorphisms were detected by PCR-RFLP,PCR-SSCP and DNA sequencing technique.Selected the genetic marker of racing performance in Yili horses,Found the association with DMRT3,MSTN and ACTN3 genes on the racing performance. Provide basis for preliminary studies and technical support for further exploration of the genetic mechanism of racing performance Yili horse breeds from the molecular level.The results were as follows: 1. Association analysis DMRT3 genetic polymorphism with gait traits in Yili horseGenetic polymorphism detection on exon 1 of DMRT3 gene in different types of Yili horse, where DMRT3 gene polymorphismin in Yili horse was the first time to report. The study found that the gene polymorphism in DMRT3, sequencing result showed that the SNP was g.22999655C> A. It was a nonsense mutation that created a premature stop codon that results in a truncated protein. There were three genotypes : CC, CA and AA, C was the dominant allele. After association analysis,Speed of pacing type was faster than trotting type(P<0.05).the SNP was not associate between DMRT3 gene mutation and gait type in Yili horse, but it had a significant effect(P <0.05) of speed in Yili horse(Trotting type) and may be an important site on gait traits of Yili horse. 2. Association analysis MSTN genetic polymorphism with medium-long distance in Yili horseGenetic polymorphism detection on intron 1 of MSTN gene in 4 groups of Yili horse.The study found that the gene polymorphism in MSTN, sequencing result showed that the SNP was g.66493737C> T.There were three genotypes : CC, CT and TT, C was the dominant allele in 2km and 15 km group,while T was the dominant allele in 3km and 5km group. After association analysis, in 5km group,this mutation had a significant effect(P <0.05) of speed in Yili horse and may be an important site on middle-long distance of Yili horse.3. ACTN3 genetic polymorphism detected in Yili horseGenetic polymorphism detection of ACTN3 gene on different types of Yili horse.There were no polymorphisms in 10, 11 and 11 exons of the gene. |
