In this experiment,eleven kinds of Chinese herbal medicine(Codonopsis pilosula,Largehead atractylodes rhizome,Poria cocos and Liquorice etc.)were processed into supplementary SIJUNZI decoction(SJZD)and added into the diet of piglets in order to study the mechanism of supplementary SJZD on effect of immune function in piglets.Sixty crossbred piglets “Duroc × Landrace × Yorkshire”(1-day-old)were selected from 6 litters and randomly divided into three groups,two litters of 20 piglets each.Piglets in control group were fed with milk replacer,piglets in 1% supplementary SJZD group and 2% supplementary SJZD group were fed with milk replacer plus 1% and 2% supplementary SJZD from 14-day-old,respectively.All the piglets were weaned at 21 d.The trial was conducted for 14 d.Blood samples of ten piglets from each group were taken from vena cava anterior at 14,21 and 28 d,and the sera were separated for measuring the levels of IFN-?,IL-2,IL-4 and IL-6.Meanwhile,the piglets were weighed to calculate the average daily gain(ADG),average daily feed intake(ADFI)and the ratio of feed and gain(F/G).At 28 d,six piglets from each group were slaughtered,and the small intestinal mucosa was collected and stored at-20 ?for measuring sIgA levels.Each segment of the intestine was also collected and placed in 4% paraformaldehyde and liquid nitrogen for determining the positive expression and distribution of sIgA,IgG and IgM,and the relative expression of sIgA,IgG and IgM mRNA,respectively.The results were as follows:1.Compared with control group,at 21 d,the average body weight(ABW)of piglets in 2% supplementary SJZD group was significantly increased(P <0.05);at 28 d,the ABW of piglets in 2% supplementary SJZD group was higher than those of 1% SJZD supplementary group and control group(P <0.01),and the ABW of 1% supplementary SJZD group was higher than that of control group(P <0.05).Compared with control group,at 14-21 d,the ADG of piglets in 2% supplementary SJZD group was significantly increased(P <0.05);at 21-28 d,the ADG of piglets in 2% supplementary SJZD group was higher than those of 1% supplementary SJZD group(P <0.05)and control group(P <0.01),while the ADG of 1% supplementary SJZD group was higher than that of control group(P <0.05).At 21-28 d,ADFI of piglets in 2% supplementary SJZD group was greatly significantly higher than that of control group(P<0.01).F/G of piglets in supplementary SJZD groups was lower than that of control group,and the F/G of piglets in 2% supplementary SJZD group was lower than that of 1% supplementary SJZD group,but there was no significant difference among groups.2.At 28 d,compared with control group,serum IFN-? levels of piglets in 2% supplementary SJZD group were significantly increased(P <0.01).Serum IL-2 levels of piglets in 2% supplementary SJZD group were significantly higher than those of control group(P <0.01),and 1% supplementary SJZD group was higher than control group(P <0.05).Serum IL-4 levels of piglets in 2% supplementary SJZD group were significantly higher than those of control group(P <0.05).Serum IL-6 levels were significantly higher than those of 1% supplementary SJZD group(P <0.05)and control group(P <0.01),and 1% supplementary SJZD group was higher than control group(P <0.05).3.Compared with control group,the intestinal mucosal sIgA levels of piglets in supplementary SJZD groups were significantly increased(P <0.01),and sIgA levels of piglets in 2% supplementary SJZD group were higher than those of in 1% supplementary SJZD group(P <0.01 or P <0.05).4.sIgA,IgG and IgM positive expression in intestinal tissues of piglets in 2% supplementary SJZD group were significantly increased than that of control group and 1% supplementary SJZD group(P <0.01 or P <0.05).5.Compared with control group,sIgA,IgG and IgM mRNA relative expression values in intestinal tissues of piglets were significantly increased in 2% supplementary SJZD group(P <0.01)and were increased in 1% supplementary SJZD group(P <0.01 or P <0.05).Meanwhile,sIgA,IgG and IgM mRNA relative expression values in intestinal tissues in 2% supplementary SJZD group were higher than those of 1% supplementary SJZD group(P <0.01 or P <0.05).In conclusion,supplementary SJZD cauld improve growth performance,and enhance the intestinal mucosa immune function of piglets.The Effect of 2% supplementary SJZD were prior to 1% supplementary SJZD.
