Milk thistle Silybum marianum(L.) Gaertn. Also known as silymarin pheasant, milk thistle, Acanthus ilicifolius, Silybum marianum plant of the genus dried ripe fruit is used as medicine. Native to southern Europe, North Africa, in Europe as a folk medicine for the treatment of hepatobiliary disease history of 2000 years. In 1952, the Beijing Botanical Garden from the United Kingdom introduced as an ornamental plant cultivation, 1972 by the China National Native Produce and animal by products import and Export Corporation, Tianjin native branch from Germany introduced as medicinal plant cultivation. The study found that silymarin has powerful antioxidant function, can protect liver cells from free radical damage, effect is far better than vitamin E; promote protein synthesis, accelerate the manufacture of new liver cells, or order of the damaged liver cells to repair itself. It is called "natural hepatoprotectants". In addition to the prevention and treatment of liver and gallbladder disease, but also has antioxidant, anti radiation, anti-tumor, improve the immunity and other functions. Silymarin has been FDA as a drug treatment and nutritional intake of liver toxicity included. Besides medical value, silymarin is widely used in oil, feed, honey, ornamental and health food etc..The Silybum marianum Gaertn roots as explants, Agrobacterium rhizogenes A4 bacterium infecting induce hairy roots and screening culture medium, inoculation amount, temperature, nitrogen source, phosphorus source, hormones and different elicitors on accumulation of Silybum marianum hairy root growth and silybin influence. Experimental results revealed that the kinds of culture medium, inoculation amount, temperature, nitrogen source, phosphorus source are the accumulation of hairy root growth and silybin influential. The most suitable culture conditions for MS liquid culture medium, inoculation amount is 0.2g, the culture temperature is 26 degrees C, NH4+/NO3- is 1:2, the amount of KH2PO4 is 2 times.The accumulation of IBA, 2,4-D, NAA, 6-BA, IAA, KT and adding growth of silibinin on hairy roots of different. NAA, 6-BA, IAA, IBA four kinds of hormone promotes the silymarin hairy root growth and secondary metabolites. The results of orthogonal experiment showed that the most suitable hormone was NAA, IBA, IAA, 0.5mg/L, 0.2mg/L, 0.5mg/L, respectively. Elicitor on hairy root growth and silymarin silymarin of different accumulation have different effects. Ca2+, Fe2+ for the MS culture medium provisions for the best. Salicylic acid and coniferyl alcohol, Ag+ can improve the yield of silymarin. Salicylic acid is 50 g/mL, the hairy roots of multiplication was 24.02 and the yield of silymarin 2753.35 G. Coniferyl alcohol is 30 g/mL, silybin yield is 2493.07 G. Ag+ is 10 g/mL, the proliferation rate is 16.38, the yield of silybin was 3591.01 G.
