Analysis of karyotype of ten species Elephantgrass in Fuzhou Fujian province was conducted,and the reaction procedure of RAPD-PCR and ITS-PCR was optimized. And then their genetic relationship and genetic diversity were studied.(1) According to the morphological index of JunCao such as stem diameter and plant height and so on,10 material could be divided into two categories:the one were Jujuncao,GuiMinYin, purple vanilla, hybrid Pennisetum, Zhangzhou vanilla and Pennisetum purpureum, which had the thicker stem, higher plant height,the more width and length leaf and the Jujuncao and imperatoria were the most obvious; the other were Fibrous elephantgrass, GuiMingYing and LaiMu, which had the shorter plant height, the thiner stem and narrower leaves.(2) Karyotype analysis suggest that all ten Elephantgrasses have twenty-eight chromosomes in accordance with Pennisetum purpureum Schum, the Jujuncao's karyotype was 2B, which belonged to the symmetrical karyotype and was relatively primitive species in evolution.(3) Six RAPD primers used for RAPD analysis were screened from 36 RAPD primers.33 sites were detected in the six RAPD primers, including polymorphic loci was 100%, Nei's 0.3175 due to various degrees, Shannon information index was 0.4933. This marker showed the Elephantgrass have abundant genetic polymorphism.(4) The ten Elephantgrasses were divided into two groups by the cluster analysis of RAPD. The result indicated that the Jujuncao had close relationship with imperatoria and elephantgrass of ZhanZhou, but had far relationship with Fibrous elephantgrass, GuiMingYing and LaiMu.(5) The analysis of ITS sequence showed that the size of ITS sequence is approximate 600bp,Purple Elephantgrass, GuiMu 1, Hybrid penisetum, Elephantgrass of ZhanZhou, Iimperatoria has the high homology with Jujuncao,99.7%,99.4%, 98.0%,99.1%,98.5% respectively.(6) The phylogenetic analysis about ITS sequence indicated that the ten Elephantgrasses were divided into two groups. the one were Imperatoria, elephantgrass of ZhanZhou, Hybrid penisetum, GuiMu 1, Purple Elephantgrass and Jujuncao;the other were Elephantgrass, LaiMu, GuiMingYing and Fibrous elephantgrass...
