Liuzhou Jiuwan Mountain located in northern Guangxi Rongshui, Huanjiang and Loucheng and other junction of three counties, the unique geographical and climatic conditions suitable for the growth of tea, tea is rich in wildlife resources, but the study is not aimed at the local wildlife resources.In this study, Liuzhou Jiuwan Mountain wild tea tree resources as materials,93 parts of tea plant material selected preliminary morphological markers, the use of ISSR Liuzhou Jiuwan Mountain wild tea resources and genetic diversity and relationship were studied. UV spectrophotometric analysis and HPLC analysis tools, analysis and detection of which 51 per plant for two years in the spring, summer, autumn shoots water extraction, tea polyphenol, amino acids, alkaloids and catechin components and content, amino acid composition and content of the main results obtained are as follows:1. Genetic diversity and relationship:10 amplified bands were clear, highly polymorphic, reproducible ISSR primers 93 Liuzhou Jiuwan Mountain wild tea plant and six control tea varieties (lines) of the genetic diversity and genetic Relationship analysis. A total of 112 bands were amplified, of which 105 were polymorphic, accounting for 93.75 percent, indicating Liuzhou Jiuwan Mountain wild tea populations genomic DNA with high polymorphism. Similarity coefficient between the test material is from 0.23 to 0.82, with an average of 0.51. Conducted using UPGMA cluster analysis showed that the average similarity coefficient (0.51) at the test plant copolymer of 10 groups, including three compombination group, seven independent groups. The results showed that:Liuzhou Jiuwanshan wild tea plant populations most close genetic relationship, indicating that the genetic relatively stable, while its genomic DNA with high polymorphism, indicating that the resource has a rich genetic diversity, such as protection and utilization of the resources to provide a scientific basis2. Biochemical component analysis:Through the main biochemical components in Liuzhou Jiuwan Mountain 51 wild tea tree resources in two quarters six samples were analyzed, the water extract content mean of 40.79%,27.25% polyphenols mean, mean total free amino acids was 3.12% phenol ammonia ratio mean of 8.73, theobromine, caffeine, gallic acid mean were 2.40mg/g,4.24% and 1.79mg/g. On the whole, indicating Liuzhou Jiuwan Mountain wild tea is an excellent resource of local resources, it has a high water extract, high tea polyphenols, a higher content of free amino acids and alkaloids characteristics. Its phenol ammonia ratio mean of 8.73, indicating that the resource has a characteristic black and green system, which can show an excellent selection of black and green tea varieties.Analysis showed that specific differences:the coefficient of variation Liuzhou Jiuwan Mountain wild tea resources caffeine is 11.56%, performance is more stable, and theobromine, gallic acid variation coefficient of two substances were above 30%, the larger variation range. The average sample catechin component content of 6 times for two years, three quarters of EGC (10.23 mg/g), D, L-C (7.91 mg/g), EC (4.44 mg/g), EGCG (60.01 mg/g), GCG (62.27 mg/g), ECG (21.75 mg/g); the difference between the tea catechin component content of 51 parts per plant large amplitude, three quarters of the average coefficient of variation of two EGC (35.00%), D, L-C (32.49%), EC (36.49%), EGCG (14.86%), GCG (15.83%), ECG (18.80%).15 quality trait indicators are divided into five principal components factor, the cumulative contribution rate of 83.36%, the first principal component factor has the largest contribution of 25.36%.The total amount of amino acids in the spring, EGC, gallic acid content is high.The content of water extraction?tea polyphones?phenol ammonia ratio,and the alkaloid is low The summer amino acid substances, EGCG content, catechin, ester low levels of catechins, water extracts and amino phenol is relatively high. The high autumn tea polyphenols, catechins ester ratio remaining low-quality performance of the chemical composition of the center. Each quality different chemical composition there are some significant differences between seasons, which provides a reference for the scientific use of resources.The results show that screening from Liuzhou Jiuwan Mountain spring wild tea tree resources in the biochemical composition of 7 (5,23,24,26,30,34,52)high-quality resources, together showed a higher content of flooding, moderate polyphenols, the concent of amino acid more than 3%, phenol ammonia ratio(8-15), the concent of caffeine more than 5%, the concent of catechins and EGCG is moderate:a screened with high tea polyphenols (> 34%) properties.It can be used as high-quality green and black tea in the future breeding of the plant; while filtering out 1 (No.27) having a high polyphenol (> 34%) plant characteristics, one (No.79) having a high amino acid (>4%) characteristics of plants,5 plants (4,5,12,26,34) having a high caffeine (>5%) properties,10 plants (4, 27,34,40,42,46,53,79,87,91) having high catechin (>plant 180mg/g) characteristics, one(No.91) having a high EGCG (> plant 80 mg/g) characteristics in the future be based on market demand development.3. Amino composition analysis:Among the tested plant,18 kinds of amino acid composition is relatively complete category, with higher levels vary widely, the overall coefficient of variation is large, many types of variation can provide rich material for the breeding selection; Theanine, aspartic acid, serine, histidine and glutamic acid are higher levels of five amino acid components, a large number of these amino acids account for about 60% of the total amino acids. Theanine, aspartic acid, serine, alanine and glutamic acid is correlated with the total amino larger five components. Correlation analysis showed: between a large number of amino acid and total amino acid content was significantly correlated not only the content of theanine significantly correlated and highly significant correlation with the total amino acids (TAA) and a large number of amino acid content (LAA) exist. Liuzhou Jiuwan Mountain screened from wild tea tree resources in 7(3, 7,10,12,13,70,79) high theanine content in terms of resources,9 parts (10,15,23,29,35, 49,44,76,79)of a high essential amino acid content resources,providing a reference for future breeding...
