| Bletilla striata is Orchidaceae Bletilla perennial plants, with good ornamental value and medicinal value. Due to the long-term man-made predatory excavation and lack of protection, wild resources of Bletilla striata reduce day by day. Bletilla striata medicinal materials to appear serious shortage, has been listed as one of China 30 kinds of rare endangered natural medicine. At present, people usually use division propagation to breed Bletilla striata, but the growth cycle is long, low propagation coefficient, it is difficult to meet the market demand, and plant tissue culture seedling method can obtain a large number of seedlings in a short period of time. So the use of plant tissue culture techniques were employed to study the efficient and fast breeding methods of Bletilla striata future research and protection of wild resources utilization and development has important significance.This study used wild Bletilla striata seeds as experimental material, systematic study on the key technology of every link in the tissue culture and seeding hardening and transplanting of Bletilla striata, successfully established efficient micropropagation system of Bletilla striata. The following results were obtained by experiments:1. Culture conditions:temperature 20℃,1500-25001x light intensity,10 hours illumination everyday.2. The best seed disinfection method:Put the seeds stained with detergent in the water flushing 15min. and then in the super clean bench with soaked in 75%alcohol for 30s, washing 1 times with sterile water, soaked in 0.1% mercuric chloride 15min.Finally, washing four times with sterile water.3. The germination rate of all kinds of culture medium was above 80%, the best germination medium was MS+0.1mg/L NAA, the germination rate was 100%, and played a certain degree of proliferation.4. Multiplication culture of the best training base is MS+1.5mg/L 6-BA+0.5 mg/L NAA, proliferation coefficient was 2.2, multiplication coefficient reached and seedlings grow well, plant fresh and green.5. The suitable medium for rooting was:MS+1.0mg/L GA3+1.0 mg/LNAA. average rooting number was 4.6, average root length was 2.0cm, and length of root stout uniform, low death rate, leaf fresh and green, growth vigorous.6. Transplanting:plantlets for 6 days, transplanting matrices for vermiculite:peat: Perlite=1:2:1, is the most suitable transplanting matrices and seedling survival rate can be reached 90.6%. |
