Study On Breeding Technique Of Bletilla Striata

Bletilla striata,orchidaceae,bletilla,is a perennial herbaceous plant and belongs to ground-living orchid.It has high economic and medicinal value.The growth rate of Bletilla striata is slow,the general rate of tubers is low and the cost is high,seed germination routine is easy,but the germination rate is very low,all of them makes the seedling reproduction of Bletilla striata have not been further applied.With the application and development of tissue culture in Orchidaceae,the rapid propagation technology of Bletilla striata has been rapid development.However,there are few reports about seed germination of Bletilla striata seed.In this experiment,we used Bletilla striata seeds as materials,optimized the cultivation conditions of Bletilla striata and explored the direct seeding breeding technique of Bletilla striata.We initially established a system of rapid propagation technique of Bletilla striata and provided technical support and theoretical guidance for Bletilla striata standardization and factory production.The experimental results are as follows:1.According to tissue culture experiment and aseptic germination of Bletilla striata,we got the best culture conditions of germination and growth in each stage:?1?The optimal medium composition under light are as follows:hyponex No.1+30 g/L sucrose+7 g/L agar+50 g/L banana juice+0.5 mg/L 6-BA+1.0 mg/L NAA+1 g/L activated carbon?pH=6.5?.?2?The best culture condition of protocorm subculture differentiation medium:1/2MS+1.0 mg/L 6-BA+0.5 mg/L NAA+50 g/L kiwifruit juice?pH=6.5?,and the effect of three factors is additives>NAA>6-BA.?3?The best condition of Bletilla striata seedlings subculture:hyponex No.1+2 medium supplemented with 30 g/L sucrose+7 g/L agar+0.5 mg/L NAA+2 g/L activated carbon?pH=6.5?.?4?The optimal culture condition of Bletilla striata seedling rooting:hyponex No.1+2medium supplemented with 7 g/L agar+0.5 mg/L NAA+50 g/L banana Juice?pH=6.5?,carbon source was sucrose,and it is better not adding activated carbon.?5?The best condition of transplanting seedlings:Bletilla striata seedlings with corms were used for transplanting,and planted by cluster way,2-3 strains for one cluster,The ratio of seedling to substrate is nutrient soil+perlite+peat?9:1:9?.2.The optimum culture conditions for seedling of Bletilla striata:The light intensity is2500 lx,humidity is 85%-90%,the temperature is 27?.The number of seedlings per unit is highest and up to 9 plant·cm^-2 under the treatments of 9:1:1 substrates in volume?nutrient soil:vermiculite:perlite?,sterilization by potassium permanganate and seed dressing by acticarbon in volume?seed:acticarbon=1:30?.Bletilla striata seedlings grew better under the treatment of spraying 1/2MS nutrient solution every 30 days and meanwhile adding exogenous hormone 6-BA?1 mg/L?.
