Salmonella enteritidis are not only a threat to the poultry industry and a major pathogen dangers,but also an important foodborne pathogens likely to cause food poisoning,caused by human intestinal diseases,endangering human health and development.Livestock and poultry products,eggs,meat are an important source of infection of Salmonella enteritidis and Salmonella enteritidis has a wide range of transmission,including humans widely susceptible animals are an important cause more difficult to prevent and control Salmonella enteritidis.Genome-wide association study(GWAS)was screened in the genome-wide analysis of the impact of high-performance genetic variation,with high statistical power,and high precision and widely used in important economic traits and complex study of human disease genetics of animals.In this study,Salmonella enteritidis infections 200 Jining Bairi chickens using case-cntrol experimental design,and using 600 k chicken high density chip of cecal Neishamenshi bacteria content difference larger for genome-wide association analysis(GWAS),Screening and in genome-wide Salmonella enteritidis infection-related genes or molecular markers by real-time quantitative PCR,further analysis of the expression of different genes and genotypes of each relevant Salmonella enteritidis Infection.Through GWAS analysis were detected three SNPs and Salmonella enteritis bacteria infection significantly associated,which are located in three different genes,which rs313644723(C/T)was located on chromosome 1,Wnt7b(Wingless-type MMTV integration site family)was located upstream of rs313644723(C/T);rs80757564(G/T)was located LRP5 low density lipoprotein receptor-related protein 5 gene in paragraph 11 introns of chromosome 5;rs313281555(A/G)was located LPP(LIM domain containing preferred translocation partner in lipoma)paragraph 7 introns of chromosome 9.Compared rs80757564(G/T)of GG genotype,TT and GT genetype was significant difference(P < 0.01),between TT and GT genotype no significant difference;rs313281555(A/G)of GG genotyped ? AA genotype and AG genotype have significant differences,between AA genotype and AG genotype no significant difference;Between rs313644723(C/T)were genotyped and differed significantly among genotype CC and TT,CT genotype,CT,and TT genotype between no significant difference,and the site and Tilquin et al.Study of White Leghorn chickens infected with Salmonella infection QTL CLOAC-ST cloacal Neishamenshi bacteria content is consistent.Fluorescence quantitative PCR results showed that there was no significant difference between in the cecum and spleen tissues,and the control group and experimental group,each candidate genes different gene type;and between individuals with different genotypes,in cecum and spleen tissues,the LPP gene different gene type no significant difference;in the spleen tissue,gene Wnt7 b,TT genotype was significantly higher than that of CC genotype(P < 0.05);LRP5 gene,G15802980 T,TT genotype and TG genotype expression was significantly higher than GG genotype(P < 0.01).Cecal tissue in the Wnt7 b gene type between there was a significant difference(P < 0.05),but there was no significant difference between the genotypes of LRP5,but the two genes had the same expression trend.To sum up,through GWAS screened the different SNPs and Salmonella enteritidis bacteria infection has an important role,respectively: rs80757564(g / T);rs313281555(A /g)and rs313644723(C / T).Three single nucleotide polymorphisms(SNPs)where the LPP gene,wnt7 b and LRP5 in chicken enteritis Salmonella infection play expression regulation.
