MRFs gene family plays an important role in skeletal muscle growth and development. As the key genes that influence meat quality and yield traits, The MRFs precisely regulate the proliferation and differentiation of myoblast. Myf6gene, a member of the MRFs family, participates in the terminal differentiation of embryonic myotube and maintains muscle phenotype in postnatal pigs. This article studied the Myf6expression at mRNA and protein levels during transcription and translation process in pigs. The Myf6mRNA and protein expression profiles in heart, liver, spleen, lung, kidney, stomach, small intestine, biceps femoris, psoas major, longissimus dorsi of Mashen pigs were constructed by qReal-time PCR and Western blotting. And on this basis, we studied the expression patterns of Myf6gene at the levels of mRNA and protein in longissimus dorsi at seven developmental stages (1,30,60,90,120,150, and180-day old) of Mashen and Large Yorkshire pigs. The main results were as follows:(1) The Myf6mRNA and protein were expressed in skeletal muscle, such as biceps femoris, psoas major, longissimus dorsi of30-day Mashen pigs, while they didn’t express in cardiac muscle, smooth muscle, and other tissues. The results indicated the tissue-specific expression of Myf6.(2) Mashen and Large Yorkshire pigs showed different expression patterns of Myf6mRNA in longissimus dorsi. During the whole detected growth stages, the relative mRNA expression of Myf6gene in Mashen pigs showed "up-down-up" trend, while there was no fixed pattern expression in Large Yorkshire. Both of these two different pigs reached the highest expression at180day of age, which were significantly higher than the other detected stages (P<0.01). The mRNA relative expression of Myf6gene in Mashen pig was greater than that in Large Yorkshire at60-,90-and150-day old (P<0.05)(3) With the increase of age, the Myf6protein expression in Mashen pig showed the downward trend, while showed "up-down" expression pattern in Large Yorkshire. They had the top Myf6protein expression at1-and90-day, respectively, which were significantly higher than other stages (P<0.01). At the same stages, the relative expression of Myf6protein in Mashen pigs was always greater than that in Large Yorkshire at all detected stages.
