Correlations Of Single Nucleotide Polymorphisms In CPT2、ABHD5、ACSL1 Genes With Production Traits In FLW Cattle

Abstract/Summary:
FLW cattle is the three hybrid cattle that are successfully bred in our country.It belongs to medium beef cattle breeds. It has high growth Performance and good meat quality traits.In this study,65 Dalian Snow Dragon black cattle were taken as the research object,using PCR-RFLP marker method combined with DNA sequencing to detect the PolymorPhism analysis of CPT2, ABHD5 and ACSL1 gene. The PurPose of this study is to exPlore the influence of CPT2, ABHD5 and ACSL1 gene to beef Production and meat quality traits,in order to Providing certain theoretical basis for the breeding of high quality beef cattle in China.In this study, using bioinformatics and molecular biology, the correlation analysis of relevant data were Performed between 18SNPsof the CPT2, ABHD5 and ACSL1 genes and 9 traits in 65 cattle. The mRNA exPression of three genes were determined using real time PCR at different marbling score of FLW cattleand and different tissues of shuanghe cattle’s, the results were as the following:1.Altogether 9 SNPs in CPT2 gene were detected using PCR-RFLP. The association between SNPs and carcass traits and meat quality traits were analyzed with general linear model,the result shows that:(1)The 5’upstream G78Twas associated with the fat content,beef marbling grade and fat content intramuscular,the mean of genotype GG was significantly higher than GT genotype (P<0.01); The content of fresh water of animals and fat color grade with GG genotype was higher than those with GT genotype (P<0.05).(2) The Exonl A8G was associated with weight before slautering, the mean of genotype AA was higher than AG genotype (P<0.05).(3)The Intronl G287A was associated with weight before slautering and triangle belly beef grade, the mean of genotype GG was higher than GA genotype (P<0.05).(4)The Intronl A346G was associated with weight before slautering, the mean of genotype AA was higher than GA genotype (P<0.05).(5)The Intronl G369A was associated with weight before slautering, the mean of genotype GG was higher than GA genotype (P<0.05).(6)The 5’upstream G29A,Intronl G153T,C166T,A302Gwere not significantly associated with any carcass traits and meat quality traits (P>0.05).2.Altogether 2 SNPs in ABHD5 gene were detected using PCR-RFLP combined with DNA sequencing. The association between 2 SNPs and carcass traits and meat quality traits were analyzed with general linear model, and the result shows that:(1)The 5’upstream A941C was associated with triangle belly beef grade, the mean of genotype AA was significantly higher than CC genotype (P<0.01); the fat content and fat content intramuscular of the mean of genotype AC was higher than CC genotype (P<0.05);The content of fresh water of animals of the mean of genotype CC was higher than AC genotype (P<0.05).(2)The Intron5 A204G was associated with weight before slautering and fat color grade.the mean of genotype AG was higher than AA genotype (P<0.05)3.Altogether 7SNPs in ACSL1 gene were detected using PCR-RFLP combined with DNA sequencing. The association between 7 SNPs and carcass traits and meat quality traits were analyzed with general linear model, and the result shows that:(1)The Exon6 C36G was associated with weight before slautering,the mean of genotype CC was higher than CG genotype (P<0.05);The content of upper brain cover with CC genotype was significantly higher than those with CG genotype (P<0.01).(2) The Exon7 T131A the content of upper brain cover with TT genotype was higher than those with TA genotype (P<0.05).(3) The Exon7 G132A the content of upper brain cover with GG genotype was higher than those with GA genotype (P<0.05).(4) The Exon7 C167A was associated with fat color grades,fat content,meat color grade and fat content intramuscular,the mean of genotype CA was higher than CC genotype (P<0.05); The beef marbling grade with CA genotype was significantly higher than those with CC genotype (P<0.01).(5) The Exon7 A168T was associated with fat color grades,fat content,meat color grade and fat content intramuscular,the mean of genotype AT was higher than AA genotype (P<0.05);The beef marbling grade with AT genotype was significantly higher than those with AA genotype (P<0.01).(6) The Intron5 C52T the content of upper brain cover with CC genotype was higher than those with CT genotype (P<0.05).(7)The Intron6 G101C was not significantly associated with any carcass traits and meat quality traits (P>0.05).4. There is no significant difference between the CPT2 and ABHD5gene expression in animals with significantly different marbling grade (P>0.05). The ACSL1 expression in animals with higher marbling grade is significantly higher than those with lower grade (P<0.05).5.The expression of mRNA was very significant different between the heart, liver, sPleen, lung, kidney, muscle and adiPose tissue in CPT2 gene(P<0.05),highly exPressed in liver;There is no significantly difference between the ABHD5 and ACSL1 gene expression of mRNA in the tissues (P> 0.05).
