Mx(Myxovirus resistance) is one of antiviral protein family members generated by inducing host cell by type Ⅰ interferons (IFNα/β), with the biological activity of anti-influenza virus, vesicular meningitis virus and Thogoto virus; NRAMP1(Natural resistance associated macrophage protein,1) genes specificly expressed in phagocytic cells, such as macrophages and addicted to neutrophils and peripheral blood cell, and related to Salmonella and parasitic pathogens resistant to multiple cell, which characteristic by a non-pathogenic specific; LAG-3was an inhibitory molecule, which related with maximize regulating T cell function, effector T cell proliferation, regulating the homeostasis, playing an important role in regulating the amplification of activated primary T cells and memory Tcell pool. In this study, Mx, the NRAMP1gene and LAG-3gene polymorphism were chosen as research, aiming to explore the relationship between these three genes and immune traits and carcass traits of Dagu chicken, therefor to provide a scientific basis for genetic basis to carrying out the high performance and strong disease resistance of Dagu chicken.In this study,PCR-SSCP technique was chosen to detect the single nucleotide polymorphisms of Mx gene in exon3, NRAMP1gene in Exon6and the LAG-3gene in Exon4in Dagu chicken. The results showed that:single nucleotide polymorphisms (SNPs)were detected in these sites, besides which the Mx gene in the exon3performed six genotypes:AA, BB, CC, BC, CD, AC; Exon6in NRAMP1performed three genotypes: AT, GC, AC; the LAG-3gene also showed the three genotypes CC, TT, CT.In carcass traits, relate statistical analysis showed that:to Mx gene, the carcass weight, eviscerated weight, half carcass weight, wing weight, breast muscle weight, leg muscle weight and other indicators of individuals of genotype CC are lower than other genotypes individuals, besides which carcass weight was significantly lower than the AA, AC and CD genotype, semi-eviscerated weight was significantly lower than AA-type AB and BC individuals, wings weight was significantly lower than that of AA, BB, AC, CD and BC individuals. Variants of NRAMP1gene Exon6have a certain impact on carcass traits of Dagu chicken groups, GC individuals were significantly lower than AC and AT genotype in the various carcass traits. Variation on the LAG-3gene Exon4had little effect on carcass traits of Dagu chicken. Carcass weight of CC genotype Individual was significantly higher than the individual genotype of TT, also higher than CT individuals, but the difference was not significant.In immune indicators, statistical analysis results showed that:AT individual of NRAMP1gene Exon6point, the total number of leukocytes was significantly higher than AC and GC genotype; while the different genotypes of other genes had little effect on the total number of white blood cells and lymphocyte counts in several immune indicators. In the blood biochemical parameters, correlation statistical analysis results showed that:LD content of Mx gene of AA-type individual was significantly higher than the BB,CC, AC, BC-type individual,but the others were not significant; LD content of NRAMP1gene AT-type individual was significantly higher than the GC genotype and AC genotype, on the other traits did not find a significant effect; LD content of AT genotype was significantly higher than the GC and AC, the other characters had no significant difference in performance. The relationship of different genotypes and carcass traits of NRAMP1gene Exon6could speculat that the variation had great significance in the improvement of productivity and disease resistance of Dagu chicken.
