Achyranthes bidentata polysaccharides (ABPS)receives great attention in recent years with its extensive biological activity. It is reported that ABPS has muti-roles in regulating cell proliferation, differentiation and apoptosis, and especially in activation of immune cells. What's more, ABPS shows many biological activities, such as anti-tumor, anti-virus, ant-oxidation, enhancing immunity, anti-diabetics and anti-aging. However, the effect of ABPS on intestinal epithelium cell was unclear. This study explored the effect of ABPS supplementation on proliferation of intestinal porcine epithelial cell line (IPEC-J2) and immunoregulatory function of ABPS on LPS-challenged cells.In order to explore the effect of ABPS on cell proliferation, five groups of IPEC-J2 received different dosage of ABPS administration, Group A (0 ?g/ml ABPS), Group B (300?g/ml ABPS), Group C (600 ?g/ml ABPS), Group D (900 ?g/ml ABPS) and Group (1200 ?g/ml ABPS), and processed cell accounting at 24h,48h and 72h, respectively. While, the same dosage of ABPS was used to study the immunoregulatory effect of ABPS on LPS-challenged IPEC-J2.The results shown that ABPS with dosage of 1200?g/ml significant promoted the proliferation of IPEC-J2 both in 24h (P<0.01) and 48h(P<0.01) than other groups,and ABPS at dosage of 1200 and 900 ?g/ml could both significant promote cell proliferation than other groups (P<0.01).In LPS-challenged cell, ABPS with lower dosage administration significant promoted the expression of TLR4 both on mRNA and protein level, while high dosage of ABPS shown little effect on TRL4 expression. Whit the increase of ABPS dosage, the gene expression of mitogen activation protein kinase (MAPK) grads increased,while on protein level, the expression of MAPK was inhibited with lower dosage of ABPS (300 ?g/ml and 600?g/ml) but was not significant affected in high dosage;the administration of ABPS significant active the phosphorylation of MAPK. And lower dosage of ABPS (300?g/ml)shown significant repressive effect on nuclear factor kappa B (NF-?B) pathway. ABPS supplementation significant decreased the concentration of interleukin-1,2,6,8 and tumor necrosis factor alpha in the supernatant of cell culture media, and shown a negative correlation with the dosage of ABPS. Collection, ABPS could promote the proliferation of intestinal epithelium cell, and could remised the injury induced by LPS challenge by activation of MAPK pathway and inhibition of NF-?B signaling way, and decrease the concentration of pro-inflammation cytokines.
