In the process of daily inspection and quarantine,there are six kinds of bean-like insect pests with similar morphological features.The coffee beans(Araecerus fasciculatus(De Geer)belongs to Anthribidae.Callosobruchus maculatus(Fabricius)Caryedon serratus(Olivier)Acanthoscelides obtectus(Say)C.analis(Fabricius)C.chinensis(Linnaeus)belong to Bruchidae.And A.fasciculatus?C.maculatus?C.serratus ?A.obtectus?C.analis are in the entering prohibited list of quarantine pests.C.chinensis is the quarantine pest in the Sino-Russian,South China phytosanitary plant protection agreement.The economic significance of the six kinds of beans are very important,the six kinds of beans are mainly identified by the external morphological characteristics of adults,however the morphological characteristics are very similar causing false check.In addition,the port intercepted not only adults,also some eggs,larvae,pupae and other immature insects,These six kinds of beans in the port was intercepted frequently,it of important guiding significance to find out a correct and rapid identification method in the port inspection.In this study,the mitochondrial gene of four peanuts was extracted from the four cultivars,and the mitochondrial specific gene Co1 of six bean elephants was extracted and sequenced by design primers.And then the molecular identification of the mitochondrial genome and the further analysis of the mitochondrial genome sequence were carried out.It will be conducive to the establishment of a more robust phylogenetic relationship.The results are as follows.(1)The specific gene Co1 of the mitochondrial DNA was extracted from the bean image of the Linyi Entry-Exit Inspection and Quarantine Bureau and the bean image in the Tai'an grain market sedimentary storage(beans)Identification sequencing.(2)Co1 gene was 420 bp in length and Blast was used to analyze the difference of Co1 gene.(3)Based on the Co1 sequence of four kinds of experimental material,the phylogenetic tree of different bean elephants found in Gen Bank was used to analyze the phylogenetic tree.The phylogenetic relationship of different bean elephants was analyzed,and the reference system was constructed.The insects were identified by molecular sequencing conveniently.(4)Validation: The individuals were identified and sequenced into the phylogenetic tree by interception of quarantine eggs,such as adults,and sequenced.
