In this study,the test materials including Acanthoscelides obtectus (Say),Callosobruchus maculatus (Fabricius)and Callosobruchus chinensis (Linnaeus) belong to pest of Bruchidae.Their morphological characters are very similar. Within them, Acanthoscelides obtectus (Say)and Callosobruchus maculatus (Fabricius) have been identified as quarantine pest by government. Callosobruchus maculatus (Fabricius) was a regulated quarantine pest on the Bi-agreement of plant qurantine and protection of Sino-Russia and Sino-Yugoslavia. The economic significant of these three pests is very important. Bruchidae pest are mainly identified by means of morphological characters of adults in port administration of inspection and quarantine, but it is easy to misidentify because of their very similar morphological characters.Primers were designed and synthesized based on the sequences of species-specific mitochondrial Cytb genes in Acanthoscelides obtectus (Say),Callosobruchus maculatus (Fabricius),Callosobruchus maculatus (Fabricius). DNA extracted from them were used as templates of PCR amplification. The gel electrophoresis patterns of PCR products were applied as molecular markers to identify these three pests.The molecular identification technique system was successfully established to supply molecular biological efficient, stable, exact and economic method for quarantine of these three pests.
