The experiment was conducted at the College of Horticulture Science and Engineering of Shandong Agricultural University and the National Research Center for Apple Engineering and Technology from March 2015 to May 2017.The effects of different disinfection treatments on the contamination rate,sprouting of axillary bud and survival rate of explant in raspberry tissue culture were studied by using the stem section of ‘Tulameen' Rubus corchorifolius L.f.as the material.The initial medium and the subculture medium were optimized to obtain the most suitable addition ratio and concentration of hormone by designing orthogonal experiment in the process of raspberry tissue culture.The aim was to provide guidance for the rapid propagation of raspberry.The main results were as follows:1.The best method of disinfection for explants of raspberry tissue culture was treated with 75% alcohol for 20 s and then treated with 1% sodium hypochlorite for 20 min.It can decrease the contamination rate by 20%,increase the rate of sprouting of axillary bud to 60%,and the survival rate to 100%.It can not only effectively prevent pollution,but also effectively improve the rate of sprouting of axillary bud and the survival rate of the explants.2.During initiated culture,under the condition of MS+ 6-BA 1 mg / L +NAA 0.5 mg / L +30 g / L sucrose and MS +KT 2 mg / L +NAA0.2 mg / L +30 g / L sucrose,it can result in the higher average plant height and the average number of leaves.3.In the process of proliferation culture,under the condition of the MS +6-BA3 mg / L +NAA 0.1mg / L+ IAA 0.3mg / L +30 g / L sucrose,it could result in average proliferative number of 4.5 and the average plant height of 1.65 cm.4.In the process of rooting culture,the rooting rate was 91%,the average root length was 3.0 cm,and the average root number was 3.8 under the condition of 1/2 MS +6-BA0.5 mg / L +3% sucrose +0.8% agar which can effectively improve the quality of rooting.5.The seedling survival time could affect the seedling survival rate,and the seedling survival rate could reach 80% at 30 days.
