| This paper focuses on screening the best explant of red raspberry Heritage, to research the influence of plant growth regulators on red raspberry buds, subculture and rooting,and filter out the best recipes for each stage, transplanted successfully the last. Meanwhile research raspberry plantlets browning control strategy, trying to find effective measures to control all stages of red raspberry tissue culture browning. The main findings are as follows:1.Establishment of Red Raspberry tissue culture systemMake a complete randomized trial on Explants acquisition time, bud type and sterilization time. Tests show that non-nude bud for the best explants, suitable for non-nude shoots sterilization time is 8min. In bud induction cultured experiments,Through the 6-BA, GA3 and NAA orthogonal analysis, screen out the best suitable formulation of red raspberry bud induction is MS+6-BA1.0mg/L+NAA0.1mg/L.In the comparison of various factors,6-BA affects the most significant.By 6-BA, NAA and TDZ orthogonal analysis, screen out the best suitable formulation of red raspberry multiplication culture is MS+6-BA0.5mg/L+NAA0.1mg/L+TDZ0.5mg/L,6-BA affects the most significant.Research on the impact of basic medium types, whether they contain activated carbon,and IBA concentration on rooting. The results show the best suitable formulation of rooting is 1/2MS+0.5 g/L of activated carbon+IBA0.2mg/L。Plantlets of domesticated transplanting is a key link in the process of plant tissue culture production application, only connect effectively can maximize the advantages of plant tissue culture. This experiment based on the research of the domesticated days, shows that domestication of red raspberry plantlets before transplanting is crucial, greatly improved the survival rate of transplanting. Best to domestication in 3-5 d,and Transplanting survival rate reached 93%.2. Research on red raspberry plantlets browning control technology:By studying the different explants and sterilization methods for red raspberry browning, the results show that Segmented sterilization has obvious advantages, non-nude buds browning rate lower than bare buds and terminal bud. Therefore, the best explants of red raspberry tissue culture is non-nude shoots, sterilization time is first sterilization 4min, then sterilization 3min after rinse with sterile water. Culture in dark on browning was not significant, it can reduce the browning degree, while raspberry buds will lead to a longer time.In the subculture experiment by adding different concentrations of activated carbon, EDTA, citric acid and Na2S2O3 in the medium, derived activated carbon and citric acid on red raspberry plantlets browning control has a positive effect. Activated carbon content increases, the degree of browning continue to decrease, but the proliferation rate has also been reduced,this study thinks that in red raspberries browning controlling, the activated carbon content of not more than 2g/L. When citric acid concentration of 6g/L, the browning rate lowest, proliferation multiples of 7.17. Adding EDTA to medium not only does not reduce the browning of EDTA, but increased the browning rate and degree of browning. Adding Na2S2O3 Although will reduce the Browning rate, can aggravate the Browning degree, make the proportion of severe Browning increased dramatically.When Mg, Fe content in culture medium for 1/2 of the MS, the leaves will turn yellow, when the content of Mg, Fe promoted to MS 2 times, thick green plant, no yellowing phenomenon. But after Mg, Fe double makes Browning rate rise to 79.78%, but compared with MS there was no significant difference. Mg, Fe element can alleviate red raspberry plantlets turn yellow.The experiment to study the effects of activated carbon for rooting stage browning, when activated carbon concentration in 1.0g/L the rooting rate is higher, the higher the average root number per plant, plant root rooting fast and neat. |
