Forest musk deer(Moschus berezovskii)are important economic animal in China.In recent years,the population of forest musk deer has been sharp declining,because the musk has so great value that a large number of forest musk deers was hunted.In 2002,the forest musk deer was listed in the china animal protection at the national level.At present,the development of artificial musk deer is one of the important method to protect the forest musk deer resources,but the captive forest musk deer has brought new problems,genetic related problems are particularly prominent.At present,due to the high level of protection,the limited distribution area and the difficulty of sample acquisition,there are few reports on forest musk deer.In order to enrich the genetic data of forest musk deer and to provide information for the breeding management of musk deer,this paper has carried on the following research: Construction of forest musk deer microsatellite enrichment library by magnetic bead enrichment method and screening polymorphic microsatellites from the library.Using 8 polymorphic microsatellits to analysis the genetic diversity of the captive forest musk deer in Shaanxi and UPGMA clustering analysis was performed on 40 forest musk deer.The main results are as follows:1.The microsatellites were obtained by magnetic bead enrichment,16 microsatellites were obtained.Among them,there were 8 complete microsatellites accounting for 50 %,7 incomplete microsatellites accounting for 43.75 %,1 Compound microsatellite,accounting for 6.25 %.2.14 pairs of microsatellite primers were designed and 3 polymorphic microsatellites were found by screening.They could be used for the genetic diversity analysis of the musk deer population;An interspecific identification site of forest musk deer and alpine musk deer.3.The results of genetic diversity analysis showed that the polymorphic information of 8 microsatellites was between 0.679 and 0.870,and the average polymorphism information content was 0.798.A total of 68 alleles were detected,the average number of alleles was 8.5,and the average effective number of alleles was 5.8770.The average heterozygosity of the population was 0.5428,the average expected heterozygosity was 0.8337,and the average shannon's information index was 1.8971.The chi-square test showed that the Mber76 C loci was in accordance with the Hardy-Weinberg equilibrium and the remaining 7 sites deviated from the Hardy-Weinberg equilibrium.A total of 14 rare alleles were detected in 8 microsatellites,accounting for 20.59 % of the total number of alleles.4.The results of clustering analysis of 40 individuals indicating that the similarity coefficient of the population is between 0.72 and 0.97,the population can be divided into 9 categories,the number of same group individuals ranging from 1 to 22.This research enriched the microsatellites of forest musk deer and found that the genetic diversity of musk deer population in Shaanxi was high,but in order to improve the development of musk farmland,it is necessary to continuously improve the management level,establish a perfect population genetic lineage,real-time group genetic testing to avoid the risk of population degradation.
