E.coli is pervasive in human and animals.It is resident bacterium in intestine and also a kind of opportunistic pathogen some of the particular serotypes of E.coli are pathogenic to human beings and animals,especially infants.Rabbit diseases caused by E.coli are mixed,caused by pathogenic E.coli and its toxin,explosive naive rabbit intestinal diseases,characterized by short latent period and high mortality.All stages of age of rabbits are sensitive to E.coli.Hence significant economic loss was caused in rabbit breeding.Our experiment was carried out in order to investigating the prevalence and antibiotic resistance of E.coli in rabbit breeding farms.Twenty specimens of rabbit stools were collected respectively in three regions(Ningyang,Xintai and Dingping)in the city of Tai'an in March,2016.We collected sixty specimen in all in three regions.The collected samples were subject to selection culture medium,observation by Gram stain and isolation of E.coli by16 srRNA technique,and future chemical reaction to observe the the chemical traits of E.coli.After the isolation and identification of E.coli,K-B method was used to not only examine the sensitivity of E.coli isolates to fourteen common used antibiotics,but also know of the phenotype of drug resistance.Afterwards,the existence of twenty-four drug resistance genes in rabbit origin E.coli isolates was tested.We continued to use the method which was used to test type I integrons to investigate the drug resistance of bacteria.Finally,we performed molecular diversity analysis of rabbit origin E coli isolates by ERIC-PCR.The trial got fifty-five unduplicated E.coli isolates in total by isolation and identification of E.coli.Antibiotic sensitivity tests indicated that fifty-five isolates showed variable drug resistance profiles and sensitivity was different from one another.The resistance rate to tetracycline was highest,amount to 78.2 percent.However,isolates were sensitive to the following four drugs ceftazidime,ceftrxiaxone,imipenem and amikacin,and the resistance rates were low.The drug resistance genes detection showed that eleven antibiotic genes were found among fifty-five isolates of rabbit origin E.coli isolates,among which blaTEM andblaCTX-M were most prevalent,with positive rate 98.18 % to blaTEM and 94.5 % to blaCTX-M,followed by sul-2 with positive rate 58.18%.Type ? integrons detection shows that seventeen out of fifty-five rabbit origin E.coli isolates carried type I integrons,with positive rate 30.91%.The type I integrons detected in the erail carried seven drug resistance gene cassettes in total,which are dfrA17 + aadA5,dfrA1 +catB3 + aacA4,aadA2 + LinF,dfrA1 + aadA1,aadA22,dfrA12 + orfF + addA2,aad A16+dfrA27+arr-3,respectively.The isolates were divided into fifty-five genotype(A~W)by ERIC-PCR.,the homology between 58%-100%.Presently,the prevalent,characteristic and drug resistance are the focus of global attention.Our experiment aimed to analyze the drug resistance and integron traits of E.coli in rabbit breeding plants.We gained a brief knowledge of drug resistance of rabbit origin E.coli in the city of Tai'an,which provided reliable prilimenary basis for reasonable choice of antibiotic in clinical practice.It will contribute to wholly precaution against infection caused by E.coli.It will also helps to bring in preventive strategies.
