Lead is one of the most widely used heavy metal.Chronically exposed to lead is the major cause of lead poisning,and it often occurs in occupational disease clinically.Lead can deposit in several body parts and do harm to the tissues and organs when it invades the organism.Taurine is a kind of endogenous and damage-prevented matter with various functions,widely existing in animal body.This study chose Wistar rats as the research object,and divided them into control group,Pb group,taurine group and taurine prevention group.The study would carry out to discuss the preventive effect of taurine on lead poisoning from the following four parts after different treatment of the rats:1.The effect of taurine on growth of rats exposed to leadThe rats were divided into different groups and received different treatments,and their body weight data would be recorded weekly until the end of the study.Blood collection was implement from the abdominal aorta for plasma preparation to test the blood lead level after digested;The heart,liver,spleen,lung,kidney and testis of rats in different groups were separated to measure the organ coefficient.The results of the study showed that there was no significant difference in body weight of rats in each group(P>0.05),and lead showed a significant effect on the organ coefficient of spleen,lung,kidney and testis in rats(P<0.05),while taurine had a significant improved effect on the development of kidney and lung in rats exposed to lead(P<0.05).The blood lead ievel of rats exposed to iead was significantiy higher than those in control group(P<0.05),while the blood lead level of rats in prevention group declined significantly compared with rats exposed to lead(P<0.05).2.The effect of taurine on the injury of liver,kidney and testis in rats exposed to leadLead is accumulated in the tissues and organs as it is expeled from animal body hardly,doing damage to the tissues and organs.In this part the study mainly discussed the preventive effect of taurine on lead poisoning from histopathological changes by preparing paraffin section and ultrathin section.Optical microscopy and transmission electron microscopy examination found that the liver,kidney and testis of rats exposed to lead had different degrees of damage,while the tissue injury of taurine preventive group rats was improved after taurine prevention.The study proved that taurine could prevent the tissue injury induced by lead poisoning in rats.3.The protective effect of taurine on oxidative damage of rats exposed to lead Lead can cause the oxidation process of organism,produce ROS and damage the tissues and organs of the body mainly through oxidative stress mechanism.In this part,the study mainly discussed the preventive effect of taurine on lead-induced toxicity from the determination of biochemical indexes of tissues and blood.The isolated rat tissues and collected blood were used to make tissue homogenate and plasma,and then the detections of GSH,GSH-Px,MDA and SOD level in plasma and tissue homogenate were conducted after the sample pretreatment.The results found that the concentration of GSH,GSH-Px,and SOD in tissues and plasma of rats in Pb group were the lowest and it increased after the supplementary of taurine.But it was still lower than rats in control group and taurine prevention group I and II.The concentration of MDA in tissues and plasma of rats in Pb group were the highest and it decreased after the supplementary of taurine.But it was still higher than rats in control group ang taurine prevention group I and II,which indicated that the indexes of rats in taurine preventive group were improved to some extent.4.The effect of taurine on apoptosis of liver cells in rats exposed to lead In order to detect the function of taurine prevention on the hepatocyte apoptosis of rats with lead-induced toxicity,TUNEL method was used in this study by testing apoptosis of hepatocytes in rats.The results showed that lead poisoning can lead to the apoptosis of rat liver,and the situation of apoptosis could be improved after taurine prevention.All the results above showed that taurine had a certain preventive effect on lead poisoning in rats.
