Oxidative stress occurs throughout the life of an animal,especially during the early growth stages.Furthermore,it is a key factor that influences animal health.This study was conducted to investigate the protective effect of taurine against muscle injury induced by oxidative stress.After three days acclimatization,Thirty-five weaned and healthy piglets(9.15±0.14 kg)Duroc×(Landrace×Yorkshire)were randomly allotted into 5 groups.piglets were Intraperitoneal administered diquat to induce oxidative stress and treated with taurine(0,0.15%,0.35 and 0.6%).Sample collected at the feeding of 28 d.The results were as follows:(1)growth performance: Compared with the control group(CON),the average daily gain(ADG)and the ratio of F/G were significantly decreased(P < 0.05),and had no significant difference in average daily feed intake(ADFI)in the diquat group(DIQ).There was no significant difference in the ratio of F/G between the CON and 0.6% taurine group(HT)(P > 0.05).(2)Serum biochemistry index: Compared with the CON,the level of Interleukin-6(IL-6)and tumor necrosis factor ?(TNF-?)were significantly increased in the DIQ(P < 0.05),and had no significant difference with the HT(P > 0.05).(3)Mitochondria morphology: Expanded,vacuolated,membrane-dissolved mitochondria dissolved or faulted muscle fiber was increased in the diquat group(DIQ);the HT could reverse the morphological microstructure of muscle.(4)Free amino acids profiles: Compared with the CON,the concentration of urine arginine was significantly decreased in the HT(P < 0.05),the concentration of plasma threonine was significantly decreased in the 0.3% taurine group(MT)and HT(P < 0.05).Compared with the CON,the concentration of longissimus dorsi muscle threonine was significantly increased in the DIQ(P < 0.05),but it was significantly decreased in taurine treatment(P < 0.05).Compared with the CON,the concentration of arginine was significantly decreased but the concentration of Ile was significantly increased(P < 0.05).(5)the Metabolism of taurine: Compared with the CON,the level of urine taurine were significantly decreased(P < 0.05),taurine supplementation(0.3% and 0.6%)significantly increased the taurine level in serum,longissimus dorsi muscle and soleus in a dose-dependent manner(P < 0.05).(6)Protein degradation of skeletal muscle: Compared with the CON,the degradation ratio of protein was significantly decreased in longissimus dorsi muscle and soleus,but had no significant difference in taurine treatment,the protein expression of MAFbx and MuRF1 were significantly increased(P < 0.05)but had no significant difference in the HT.(7)Gene expression: Compared with the CON,the mRNA expression of CAT?Gpx4?SOD2?HSP70?TFAM?MAFbx?MuRF1 were significantly increased in the DIQ but had no significant difference in the HT(P > 0.05).(8)Compared with the CON,the protein expression of MAFbx and MuRF1 were significantly increased(P < 0.05)but had no significant difference in the HT(P > 0.05).In conclusion,treated with 0.6% taurine in the diet could effectively alleviate diquat-induced oxidative stress through improving the serum activity of antioxidant enzymes,elevating the concentration of taurine in tissues,restoring the morphology of muscle,remodeling the free amino acids profiles in system,recovering the expression of antioxidant genes,inhibiting the protein degradation of skeletal muscle.
