Flax is a self-pollinating herb that belongs to Linaceae,and is the main oil crop in Inner Mongolia.Flax seed is not only rich in such unsaturated fatty acid as a-linolenic acid and linoleic acid essential to human health,but also contains edible fiber,lignans,vitamins,lecithin and other ingredients,so it has the effect of lowering blood lipids,preventing cardiovascular diseases,treating cancers.The genetic diversity analysis of flax and the establishment of a core collection are of great significance to finding excellent genes in flax and flax breeding.In this paper,cluster and genetic diversity analysis of 633 flax germplasm resources were carried out using SRAP marker data and seven quality traits date,and a primary core collection was constructed.Results as follows:1.The average contents of palmitic acid,stearic acid,oleic acid,linoleic acid,linolenic acid and lignans in flaxseed were as follows:6.13%,5.76%,27.77%,12.51%,47.8%,7.23%.The average coefficient of variation(CV)was 0.099,ranging from 0.04 to 0.19,with the CV for oil content being the smallest(0.04),and the CV for lignans being the largest(0.19).The order of CV was:lignan>stearic acid>oleic acid>palmitic acid>linoleic acid = linolenic acid>oil content.2.An optimized 12.5 ? L SRAP PCR reaction system was established.Twenty pairs of SRAP marker polymorphic primers with clear band and good repeatability were selected for subsequent analysis.These primers pairs generated a total of 264 bands,of which 145 bands were polymorphic with the polymorphism ratio of 54.93%.The number of amplified polymorphic bands for each pair of primers was between 4 and 11,and the polymorphism information content of each pair of primer ranged from 0.21 to 0.59,with an average of 0.43.3.A primary core collection of 149 germplasms were selected.The average number of the quality traits of the initial core samples is similar to that of the total samples.The variance of the quality traits is larger than the variance of the original population.The primary core germplasm can effectively represent the whole flax population.After the cluster analysis,when cluster distance was 0.42,the flax germplasm resources can be divided into three categories,when cluster distance was 0.33,the flax germplasm resources are divided into five sub-categories4.In this core collection,the genetic basis of the flax resources from different geographical regions was relatively rich;and the genetic similarity coefficient of Hebei population and Gansu population was the highest,0.9644,and the genetic distance was the closest.The genetic similarity coefficient of Xinjiang population and Hebei population was the lowest,which was 0.8417,and the genetic distance was the farthest.
