Polyunsaturated fatty acids in diets have a variety of roles,participate in the synthesis of fatty acids in ruminants,and participate a variety of genes and enzymes that related to lipid metabolism.The aim of this study was to investigate the effects of flaxseed on the regulation of lipid metabolism related regulators,key enzymes and related hormone gene expression in the longissimus dorsi muscle and subcutaneous adipose tissue of Yanbian yellow cattle.In this study,30 cattles were randomly divided into 3 groups(n = 10),CON group(control group),WL group and CL group feed the basal diets add 0%flaxseed,8%whole flaxseed,8%crushed flaxseed.The content of long-chain fatty acids in the longissimus dorsi muscle and subcutaneous adipose tissue of Yanbian cattle was detected by means of gas chromatograph.Real-time quantitative PCR was used to detect the gene expression of the regulatory factors(SREBP1c,PPARy,LXRa).enzyme(SCD,ACC,FAS,LPL,C/EBPa,C/EBP?,CPT1)?regulation of hormone(GHR,Ins R,GLNR,TNFa,Interleukin-6,Adiponectin)that related to lipid metabolism in the longissimus dorsi muscle and subcutaneous adipose tissue.The protein expression of SREBPlc,PPARy and LXRa in subcutaneous adipose tissue was detected by immunoblotting.The results showed that:(1)adding 8%flaxseed in diets could increase the content of n-3 PUFAs and n-6 PUFAs in adipose tissue and muscle tissue of Yanbian yellow cattle and reduce the content of saturated fatty acid(palmitic acid and stearic acid).(2)The increase of n-3 PUFAs and n-6 PUFAs in adipose tissue can promote the gene expression and protein expression of SREBP-1C and PPARy,which are involved in lipid metabolism in subcutaneous adipose tissue of Yanbian yellow cattle,and the effect of flaxseed is more obvious after crushing,(3)Increasing the content of n-3 PUFAs and n-6 PUFAs in adipose tissue is helpful to improve the gene expression of the key enzymes SCD,FAS,ACC,LPL of fatty acid synthesis in adipose tissue,the effect of flaxseed on the gene expression of C/EBPa??C/EBP? was lower than enzymes,the gene expression of CPT1 in adipose tissue was higher than in the longissimus dorsi.(4)The addition of flaxseed was beneficial to the increase of the gene expression of GHR,GLNR and cytokines(Adiponectin,IL-6,TNF?)in adipose tissues,significantly effective in adipose tissues,and the expression of Ins R was In reverse with GLNR.In this paper,(1)adding 8%flaxseed in Yanbian yellow cattle's diet was beneficial to improve the expression of C18:1,C18:2,C18:2n-6,C18:3n-3 in the longissimus dorsi and subcutaneous adipose tissue,the content of C16:0,C18:0 was reduced,and the crushed flaxseed effect was better.(2)The increase of n-3 PUFAs and n-6 PUFAs in adipose tissue,up-regulated the expression of transcription factors SREBP-1c,PPARy gene and protein expression in subcutaneous adipose tissue of Yanbian yellow cattle,and the effect of crusted flaxseed was more obvious.(3)Improve the content of flax seed in diet was helpful to improve the gene expression level of the synthesis of fatty acids related enzymes(SCD,FAS,ACC,LPL)and regulate hormone(GHR,GLNR TNF?,ADI,IL-6)in adipose tissue.
