This study aimed to investigate the effects of different microbial ecological preparations on ferme ntation quality,nutrition composition and microstructure of treated rice straw.Then we utilized nyl on bag tests to determined rumen degradation rates.The three experiments were described in this study as follows.Experiment 1: The nutrition composition and microstructure of rice strawThis study aimed to investigate the the features of nutrition composition and microstructure of rice straw.Basing on rice straw,the contents of dry matter(DM),crude protein(CP),Neutral de tergent fiber,c Acid detergent fiber,cellulose,hemicellulose and ligin were analyzed using laborato ry testing methods,and changes of microstructure were analyzed using phehol-suffufic acid colorim etry,2,2?-Bicinchoninic acid method(BCA),fourier transform infared spectroscopy(FTIR),X-ray di ffraction(XRD)analysis and eosin-methylene blue method.The results showed as follows: 1)On t he nutriment level,the dry matter content of rice straw was 25.91% FM,the crude protein content was 37.13 g/kg DM,the content of neutral detergent fiber(NDF),acid detergent fiber(ADF),cel lulose,hemicellulose and ligin were 778.42?514?438.84?264.22?75.16 g/kg DM respectively.2)On the microstructure level,the degree of polymerization of rice straw was 430,the crystalline cel lulose(1098 cm-1),C-H flexural vibrations(1375 cm-1),syringyl and guaiacyl condensed nuclei(1427 cm-1),aromatic skeletal vibration in lignin(1515 cm-1),binding force of four characteristic fun ctional groups was powerful.The crystallinity and specific surface area were 47.52% and 1.105 m2/g.In conclusion,this study had a comprehensive ananlysis in nutrition composition and microstruc ture of rice straw.Experiment 2: Effects of different microbial ecological preparations on nutrition composition and microstructure of rice straw.This study aimed to investigate the the effects of different treated rice straw silage on the ferme ntation quality,nutrition composition and microstructure.Basing on rice straw,nine treatments were set: control group(CK)?composite enzyme(C),pectinase+laccase(PL),and composite enzyme+pec tinase+laccase(CL),Lactobacillus plantarum(LP)treated group? Lactobacillus buchneri(LB)? Lact obacillus plantarum+Lactobacillus buchneri(LPB),composite enzyme+pectinase+laccase+ Lactobacill us plantarum(CLP),composite enzyme+pectinase+laccase+Lactobacillus plantarum+Lactobacillus B uchneiri(CLPB).Samples were packaged by vacuum packager in polyethylene bags,after storing at room temperature for 45 d,changes of the silage quality and nutrients were analyzed using labor atory testing methods,and changes of microstructure were analyzed using phehol-suffufic acid colo rimetry,BCA method,fourier transform infared spectroscopy,X-ray diffraction analysis and eosin-m ethylene blue method.The results showed as follows,1)On the nutriment level,compared with control group(CK),lactic acid(LA)content in enzyme preparation treatment(C,PL,CPL)was increased significantly(P<0.05),the content of pH value,acetic acid(AA),ammonia nitrogen/total nitrogen(A TN/TN),dry matte(DM)and cellulose were decreased significantly(P<0.05).The pH value,propio nic acid(PA)content,NH3-N content,crube protein content(CP),cellulose content(C)of fungus s upplymentation treatment(LP,LB,LPB)were decreased significantly(P<0.05),dry matter(DM),hemic ellulose(HC)and ligin(ADL)did not show regularity(P>0.05).The content of lactic acid(LA),acet ic acid(AA),dry matte(DM)were increased significantly(P<0.05),the pH value,ammonia nitrogen/t otal nitrogen(ATN/TN),cellulose content,hemicellulose content,ligin content,crude protein(CP)co ntent of multi enzyme fungus premix treatment(CLP,CLPB)were decreased significantly(P<0.05).2)On the microstructure level,Compared with control group,enzyme preparation treatment(C,PL,CPL)had signifcant effect on polymerization degree of rice straw silage(P<0.05),which was lower ed by 30.26%,27.11%,56.32%.The intermolecular hydrogen bond binding force was faded,enzy me preparation treatment(C,PL)did not show regularity on crystallinity and specific surface area(P>0.05).Fungus supplymentation treatment(LP,LB,LPB)had a great influnence on degree of poly merization,which were 246,216,214 respectively.The intermolecular hydrogen bond binding forc e was abated,crystallinity didn't change,whereas specific surface area increased significantly(P<0.05).The degree of polymerization of multi enzyme fungus premix treatment(CLP,CLPB)was decreas e significantly(P<0.05),reduced to 47.37%,46.59% respectively,and the intermolecular hydrogen b ond binding force was abated,which had the trend of decreasing crystallinity and increasing specif ic surface area(P<0.05).In conclusion,enzyme preparation treatment(CL)and enzyme fungus premi x treatment(CLP,CLPB)had a good effect on degradation of cellulose,these contributed to improve ment of fermentation quality and low nutrient loss,which consequently resulted into low polymera zation degree and higher specific area,Experiment 3: The effects of different microbial ecological preparations in-situ ruminal degr adability of rice straw.The present study aimed to determine nutritional value of rice straw silage treated with nine diff erent enzyme,fungus supplymentation or their combinations.Rice straw were fermented with differ ent enzymes and fungus supplymentations,and nine treatments were seted,and six Dorper × Thintailed crossbred castrated sheep were used,measured the dynamic rumen degradation rate of dry m atter(DM),crude protein(CP),neutral detergent fiber(NDF),acid detergent fiber(ADF),cellulose,hemicellulose and ligin at 0 h,12 h,24 h,36 h,48 h and 72 h.The results showed as follows: the degradation effect of enzyme preparation treatment(C,PL,CPL),fungus supplymentation treatmen t(LP,LB,LPB),multi enzyme fungus premix treatment(CLP,CLPB)were remarkably higher than cont rol group(CK)(P<0.05).The effect of different treatments were inconsistent.And enzyme preparatio n teeatment(CL),multi enzyme fungus premix treatment(CLP,CLPB)had a best effect.
