Lonicera japonica is a kind of Chinese herbal medicine with a long history,which has a very important value of development and utilization.But the honeysuckle varieties less cultivated hybrid varieties,classification is not clear,the lack of comprehensive understanding of the genetic relationship of germplasm resources of Flos Lonicerae,seriously affect the healthy development of the promotion of new varieties of honeysuckle and the whole industry,but also restricts the research of the breeding process of honeysuckle.The Kunyu Mountain honeysuckle and Haiyang natural populations of honeysuckle artificial populations as materials,using RAPD)molecular marker technique,simple sequence repeat(Simple Sequence,Repeat,SSR)amplification technology and chloroplast trnT-trnL and trnL-trnF intergenic sequence analysis technology,carry out research on the molecular biology of honeysuckle inquiry,genetic diversity among species of honeysuckle,honeysuckle variety identification,germplasm resources protection and provide theoretical basis for using.The results are as follows:The genetic diversity and phylogenetic relationship of 31 accessions of Lonicera japonica Thunb were analyzed by RAPD molecular marker technique,and the primers were amplified with the primers of the 5 primers with good repeatability and high polymorphism.The results showed that the genetic diversity and the genetic diversity of the accessions were650 bands.0-9 bands were amplified by each primer,which indicated that the genetic diversity among different provenances was higher.Using SPSS software to calculate the 31 provenances of honeysuckle genetic similarity coefficient and genetic distance,can be found in variety 1 and 31 varieties of the genetic distance between the farthest,2.507;25 and 29 varieties of species genetic distance among 0.020 recently.Based on the analysis of genetic distance,the RAPD system cluster map between 31 different provenances was obtained,and the 31 varieties were divided into the following categories.31 samples of honeysuckle germplasm genetic relation between diversity and phylogenetic analyses using SSR molecular marker,screened 5 of good repeatability and high polymorphism were detected with 36 polymorphic loci,the average of each pair of SSR primers amplified 7.2 polymorphic loci,SSR fingerprint and structure 31 strains of honeysuckle.The genetic diversity and phylogenetic relationships of Lonicera japonica Thunbgermplasm were analyzed based on chloroplast trnL-trnF sequence.The nucleotide sequence of the sequences obtained from the sequencing results showed that the trnL-trnF sequence was quite conservative in the samples,which was suitable for studying the phylogenetic relationships among the 27 samples.Dendrogram of amplified sequences of 27 samples showed that sample 20 and sample 28 and sample 9,sample 15,sample 16,sample 17 can be divided into two branches,the credibility is 64%;4 samples and 13 samples,21 samples can be divided into two branches,the credibility is 58%;7 samples and 22 samples can be divided into two a reliability of 56%.The sample 24,sample 25,sample 18,sample 11,sample 8,sample 5 can be grouped into one class,the reliability is 100%.The three methods showed that the genetic diversity was high and the genetic background was complex.Although the results obtained by different analytical methods are not completely consistent,the molecular markers have been proved to be highly efficient and stable.
