Aflatoxin(Aflatoxins,AFT)is a kind of toxic chemicals,in which aflatoxin B1(AFB1)was the highest and most toxic,and livestock poisoning caused by AFB1 has caused a serious threat on the development of animal husbandry.In recent years,curcumin has been widely used as a kind of natural food pigment,and it has been found that curcumin has certain pharmacological effects on antioxidant,anti-inflammatory and promoting digestion,enhancing immune function and other aspects.In this study,72 one-day-old AI arbor acres broilers were selected to be randomly divided into 6 groups,which were fed with blank control diet,curcumin control diet(450 mg curcumin each kilogram),AFB1 challenge group(5mg AFB1 each kilogram)and in which curcumin were fed with dietary supplementation dose of 150mg/kg,300mg/kg and 450mg/kg in the AFB1 constituted curcumin group I,curcumin group Ⅱ and curcumin group Ⅲ.The six group broilerss were fed with each kind of diet for 28 days,and the blank control diet for 7 days.Six samples in each group were taken on the 28 th and 35 th day,and the effects of curcumin on the immune organs of AFB1 sub chronic poisoning broilers were observed by histopathology and real-time fluorescent quantitative PCR.The research aims at enriching the research data of curcumin against avian mycotoxin poisoning,and providing a theoretical basis for the application of curcumin in animal husbandry.The results are as follows:Thymus: Compared with the control group,the broiler thymus index of the samples on the 28 th and 35 th day in the AFB1 control group were significantly decreased(p<0.01),nuclear debris in cortex and medulla blood significantly increased,the amount of plasma cells and karyoplasmic area was significantly lower than the polar(p<0.01)and the levels of IL-2 and IFN-γ in thymus tissue were significantly decreased(p<0.01).Compared with the AFB1 group,in curcumin group I,group Ⅱ and group Ⅲ,thymus organ index was increased,thymus histological damage were relieved to some extent,the number of plasma cells in the thymus and its nuclear area ratio were increased,and IL-2 and IFN-γ expression levels in thymus tissue were increased to varying degree on the 28 th and 35 th day.Compared with the 28 th day,the indexes of broilers thymus increased significantly in the group of curcumin on the 35 th day.Spleen: Compared with the control group,the spleen index of the samples on the 28 th and 35 th day in the AFB1 control group were significantly decreased(p<0.01),congestion of the red pulp of the spleen area was serious,the number of peripheral lymphatic lymph nodes decreased,plasma cells and karyoplasmic ratio was significantly decreased(p<0.01),and the levels of IL-2 and IFN-γ in spleen tissue were significantly decreased(p<0.01).Compared with the AFB1 group,in curcumin group I,group Ⅱ and group Ⅲ,the spleen index was increased,spleen histological injury were relieved in different degrees,the number of plasma cells and karyoplasmic area ratio in spleen tissue were increased,and IL-2 and IFN-γ expression levels in spleen tissue were increased in different degree on the 28 th and 35 th day.Compared with the 28 th day,the indexes of spleen were increased in different degrees in the curcumin group on the 35 th day.Bursa of Fabricius: Compared with the control group,the bursal of fabricius organ index of the samples on the 28 th and 35 th day in the AFB1 control group were significantly decreased(p<0.01),the cells in the lymphoid follicular medulla of bursa were sparse,the number of lymphocytes decreased,the plasma cell number and karyoplasmic area was significantly decreased(p<0.01),and the levels of IL-2 and IFN-γ in bursa were significantly decreased(p<0.01).Compared with the AFB1 group,in curcumin group I,group Ⅱ and group Ⅲ,the bursa index was increased,bursa injury were different the degree of improvement,the number of plasma cells and karyoplasmic area ratio in bursa were increased,and IL-2 and IFN-γ expression levels in bursa were increased in different degree on the 28 th and 35 th day.Compared with the 28 th day,the indexes of bursa were improved in different degrees in the curcumin group on the 35 th day.The results showed that the supplemention of curcumin in diets containing 5mk/kg AFB1,can reduce the pathological damage of thymus,spleen and bursa caused by AFB1,increase the number of organ index and plasma cells in immune organs and karyoplasmic area ratio.Also,the improvement of the expression level of IL-2 and IFN-γ AFB1 can alleviate AFB1-induced immunosuppressive effects.In this research,the effect of adding 450mg/kg curcumin on 5 mg/kg AFB1 was optimized.
