Lilium sulphureum is perennial bulbous flower and special specie of Chinese.Not only has high ornamental value and heat resistance,bulbs can also be used as food and medicine.In recent years,with the increased needs of lilies as medicinal,a large number of wild resources were excavated,and less artificial cultivation.All this reasons led to its original habitat has been destroyed.It's an endangered species endemic.In view of these questions,the author studied on introduction and cultivation,propagation and heat tolerance.In order to provide the theoretical basis for breeding new heat resistant varieties of lily from the perspective.The main conclusions are as follows:1.Lilium sulphureum was introduced into Wuhan.According to the 2-year observation,the survival rate of introduced plants in the first year was 100%,the flowing rate was 60%.The first flower bloomed on June 10 th.Individual flower bloomed for 5-7days,and the group bloomed for 21 days.Next year,80% plants can survived.Flowers bloomed since July 10 th with the flowering rate of 50%,which was delayed by 30 days than the former year.The whole growth and development period at least was 7 months.The plant is robust and the morphological traits are similar to those of the native land.That's to say that Lilium suphureum can be cultivated in Wuhan.2.The seeds,beads buds and bulbs were used to research the best breeding method.The results showed that: the seeds led to the germination rate of 70%,which were sowed in matrix(2peat:1perlite);The induction rate of small bulbs was only 22.58% under the untreated condition,the efficient differentiation was 1.71.This way was not beneficial for small bulbs growing;The beads buds of Lilium sulphureum can germinate in 7-10 days without dormancy.Over 93% of seedlings survived using the way that Growing in the matrix and then turn to the field.After 6 months,the little bulbs can increase to 1cm.In the study,we used the bulb of Lilium sulphureum as material which were cultured in MS medium supplemented with different hormone.The results showed that:The best differential callus medium was MS+6-BA 1.0mg/L+NAA 0.1mg/L+Sugar30g/L,differentiation rate was 60%.The appropriate medium for proliferation of adventitious buds was MS+6-BA 1.5mg/L+NAA 0.5mg/L +Sugar 30g/L,efficient differentiation was 2.0,there was more buds which grew robustly.The medium 1/2MS+NAA 0.1mg/L+Sugar 60g/L could help to form root and can expand the small bulb more effectively,increased 1.94,which the leaves grow better and more roots.Then transplanted into the plug tray,all of the seedlings can grow roots,and were strong.3.The results showed that different matrices and fertilizers had significant effects on plant growth and bulb yield,and interacted with each other.The optimal combination was A1B3,in this way,the germination time of the buds was concentrated,the survival rate was 70%,the plant height was medium,and the seed germination rate was 70%,and the plant height was moderate,and bulb yield significantly.4.The results showed that the heat tolerance of the plant was decreased with the prolongation of stress time.In particular,EC,Pro and soluble protein content increased by2.84 times,5.56 times and 1.48 times than the level of control,respectively.The Total chlorophyll content and SOD also decreased,by 65.75% and 36.33% at 168 h,CAT,POD decreased at the maximum after 0.5h,and decreased by 96.47% and 66.6% at 168 h.The content of MDA increased to minimum at 0.5 h(35.58% lower than that of the control)and reached a steady level at 1 h after reaching a maximum(increased 1.55 times than the control).Soluble sugar content increased first and then decreased to the control level,120 h reached the maximum,compared with the control increased by 1.37 times.Using the principal component analysis and comprehensive evaluation,the 10 original indicators can be replaced by six independent indicators,and the cumulative contribution rate can be up to 95.08%.According to the comprehensive evaluation of D value found that plant has more stronger heat resistance at 0.5h.The optimal regression equation D = 0.545-0.026X1 + 0.083X4-0.158X7 + 0.093X9 + 0.085X10,according to the equation,we can screen out the significant correlation of heat resistance to Lilium sulphureum: EC,MDA,soluble protein,CAT,POD.The results of this study can contribute to the identification and application of heat resistance of Lilium sulphureum.
