Fatty acid binding proteins 3(FABP3)and fatty acid binding proteins 4(FABP4)are important members of fatty acid binding protein family.The main role of FABPs is to regulate fatty acid uptake and intracellular transport.Although the protein sequence are very different,the genetic structure and fuction are similar.The aim of this study was to explore the effects of FABP3 and FABP4 genes on the meat traits of new varieties in new strain of Qinchuan beef cattle(Follows are abbreviated as "Qinchuan beef cattle")and to provide a theoretical basis for Qinchuan beef breeding.In order to study the effects of FABP3 and FABP4 genes on the meat traits of Qinchuan beef cattle,the biological characteristics and biological evolution information of FABP3 and FABP4 genes were analyzed by bioinformatics.We haved selected 571 Qinchuan beef cattles about 18~36 months old which were feed in the similar circumstances.And each SNP loci was alyzed which related to population genetic information and equilibrium after genotyping.The products of PCR were analyzed to search part section hereditary variation via DNAstar program and single nucleotide polymorphism of FABP3 and FABP4 genes were detected.The association were analyzed between SNP locus and the meat traits(backfat thickness,ultrasound lean muscle area and intramuscular fat).To screen the appropriate molecular markers to the breeding of Qinchuan beef cattle and the gene combinations with excellent traits were selected for the scientific breeding of Qinchuan cattle to provide theoretical support.The main results of this study are as follows:1.The FABP3 gene sequence encodes 133 amino acid while the FABP4 gene sequence encodes 132 amino acid.Bioinformatics analysis showed that the gene sequence of the family has high similarity in different species of mammalian.2.There was one SNP locus(g.60298C>T)in the third intron of the FABP3 gene.What's more,there were two SNP locus(g.2686A>T and g.2834C>G)the first intron and one SNP site(g.4220A>G)in the fourth exon of the FABP4 gene.However,there are only two genotypes which named AA and AT genotype and without TT genotype in g.2686A>T locus.3.Single nucleotide polymorphism(g.60298C>T)of FABP3 was positively correlated with intramuscular fat that the individuals with TT homozygotes had higher significantly IMF than those with CT genotype and CC genotype(P<0.05).At g.2834C>G locus of FABP4,the individuals with CC homozygotes had higher ultrasound lean muscle area than GG genotype by 7.541 cm2(P<0.01)and IMF than GG genotype by 0.254%(P<0.05)significantly,respectively.4.At g.4220A>G locus,the individuals with TT homozygotes had higher backfat thickness than CC genotype significantly(P<0.05).5.The best combination diplotype is H1H1(ACA-ACA),individuals of H1H1 had better performance than H3H3(AGA-AGA)in ULA(P<0.01)and than H3H7(AGA-TGA)in IMF(P<0.01).Thus,in this study of Qinchuan beef cattle population,it can be found that the FABP3 and FABP4 genes are closely related to the meat traits of new strain of Qinchuan beef cattle,which can provide scientific basis for Qinchuan beef breeding.
