Waterfowl breeding is well developed in Guangdong Province,but the goose plague still happens from time to time and the outbreak of epidemic may cause significant economic losses.But so far,the Goose industry is still lack of effective drugs or related biological products against GPV.For the prevention and control of the disease,it has always been based on prevention,we always immune the breeding geese before they lay eggs,which can provide protective maternal antibodies to the goslings.The serological investigation of geese and muscovy ducks can effectively help understand the level of immunization of gosling plague in Guangdong Province.This research has collected 529 serum samples from adult geese,adult muscovy ducks and goslings from the breading farms in Guangdong Province.The samples are detected by ELISA method.The results showed that there are 135 positive samples and its positive rate of GPV antibody was 25.52%.The positive rate of antibody in northern Guangdong is only 27.94% and there are 27.11%.in western Guangdong,17.74% in eastern Guangdong,25.75% in the Pearl River Delta region.Further analysis showed that 70 samples were positive among 370 samples of goose serum collected in Guangdong province,the antibody level in the goose group is 18.92%.65 positive samples were dectected from 159 muscovy ducks serum and its positive rate of GPV antibody was 40.88%.95 positive samples were dectected from 311 muscovy ducks serum and its positive rate of GPV antibody was 41.21%.40 positive samples were dectected from 218 goslings serum and its positive rate of GPV antibody was 29.63%.With the fact that the market of GPV biological products is imbalant,it’s important to develop some new preventive and protective products to control the goose plague.Firstly,this research has learnt about the gene structure of GPV,then elect the antigenic determinant of GPV and specifically amplified the VP3 antigen fragment,then the VP3 antigen fragment is linked to NoV P-particle in order to build a recombinant plasmid named P-particle-VP3.Transform the plasmid into BL21(DE3)Competent cells,then induce the expression of recombinant protein by adding gradient concentration of IPTG.The protein is best-expressed under the concentration of 1mM and the expression level reach the top in 5 hours under the concentration of 1mM IPTG.Related researchs show that NoV P-particle can be developed as a new antigen presentation platform,which can improve the efficiency of antigen presentation,enhance the antigenicity and immunogenicity.This new antigen presentation platform can result in enhanced immune response.In the meantime,the recombinant plasmids and proteins expressed in this study provide the material basis and theoretical basis for the development of the hyperimmune serum,ELISA kits and the genetic engineering vaccines.
