Alpinia zerumbet×A.henryi ‘Harvest',as a new strain from Zingiberaceae hybridization,features regular plant type,beautiful flower type,fresh color,relatively long blooming period and easy cultivation and management,and in addition,it has a high value in decoration and application.This study chooses Alpinia zerumbet×A.henryi ‘Harvest' as the study materials,and carries out a systematic study in terms of culture in vitro,saline tolerance and cultivation matrix screening.1.Culture in vitroDifferent plant sites were used as the explant materials and different disinfection methods and different hormone mixture ratios were adopted to obtain the optimal disinfection method and the best formula to generate the best efficacy at different culture stages.The sprouts the germination of which were accelerated indoors were used as the explants,the explant contamination rate was 45%,the mortality rate was 10%,and the survival rate was 45%;the best hormone mixture ratio for the induction of adventitious buds was MS+6-BA 3.0 mg/L+NAA 0.2 mg/L,and the adventitious buds inductivity was 62.5%;the best hormone mixture ratio of successive transfer culture was MS+KT 1.0 mg/L+NAA 0.1 mg/L,and the multiplying factor of proliferation was 1.73;the plants could take root without the need of rooting measures,the rooting rate was up to 100%,and the survival rate of transplantation was up to 89%.The rooting rate was up to 86%,and the survival rate of transplantation was up to 75%.Seeds were used as the explants,the contamination rate was only 6%,and the germination rate was 38%;the best hormone mixture ratio for seed sprouting was MS+6-BA2.0 mg/L+NAA 0.2 mg/L,the seed germination rate was 44%,an increase of 6% when compared with the situation of non-induction;the best hormone mixture ratio for the induction of adventitious buds was MS+6-BA 4.0 mg/L+NAA0.05 mg/L,the inductivity was up to 85.7%,and the multiplying factor of proliferation was 4.67 times.The plants could take root without the need of rooting measures,the rooting rate was up to 86%,and the survival rate of transplantation was up to 75%.2.Salt tolerance studyThe changes in growth and physiology of Alpinia zerumbet×A.henryi ‘Harvest'were studied under the stress of salt content in soil of 0‰(CK),2‰,4‰,6‰ and 8‰, the following 10 indicators were determined,and finally a comprehensive analysis was made on the salt tolerance to provide references to the actual application.Under the condition of 2‰ salt content in soil,the normal growth of the plant could be subject to a slight impact;chlorophyll,osmotic regulating substances and antioxidase activity would be unsteady somewhat at the early stage of stress,but the overall levels did not vary greatly,and the plants could become acclimated quickly to them through autoregulation.In the soil in which the salt content was 4‰,the plants presented with the relatively mild stress symptoms,.presented the changing law of first decreasing and then increasing,the remaining indicators generally presented the tendency of first increasing and then decreasing slightly,the stress symptoms could be somewhat relieved through self-regulation,but they could not recover the original level.In the environment in which the salt content in soil was 6‰,the leaves could become withered and dry,the physiological indicators became aggravated sharply,the indicators such as malonaldehyde,proline and cell membrane permeability could increase to a relatively high level at the early stage of stress and subsequently showed a tendency of constantly increasing,and the remaining physiological indicators manifested a tendency of constantly decreasing.In the environment in which the salt content in soil was 8‰,the plants manifested the more intense inadaptability,the stress symptoms were severer when compared with the processing in the environment in which the salt content in soil was 6‰,the time to become affected was advanced,and the plants could not survive in this environment.3.Cultivation matrix screeningThe changes of biomass of the plants in the soil in which salts were mixed according to different formulas were measured and the chemical properties of the mixed soil were determined to seek the formula of mixed soil suitable for the plant growth.The content of nutrient substances in the mixed cultivation matrix and the different matrix mixture ratios jointly exerted the influence on the plant growth,and the use of mixed matrixes to cultivate Alpinia zerumbet×A.henryi ‘Harvest' had a better efficacy than the use of single matrix for cultivation.The matrix mixture ratio that generated the best cultivation efficacy was: sand: turf: pond silt: yellow mud =1:3:3:3,and the increase rate in plant increment was 98.3%;...
