In this research all types of ectomycorrhizae of Picea craassifolia collected from the 21 Helan Mount National Reserve of Alashan of Inner Mongolia has been isolated under the microscope,total DNA has been then extracted.PCR has been carried out by using the primers-ITS1 and ITS4 which only exist in fungi.The ectomycorrhizal fungal diversity of Picea crassifolia has been analyzed after the operations including quality control of sequcnces,sequence alignment.OTUassignment.analysis of diversity,classification,as well as phylogenetic analysis.The results showed that P.crassifolia has high diversity on ectomycorrhizal fungi,268 representative sequences of OTUs has been obtained,The average sequence length is 241 bp,these OTUs were belonging to 3 phylums,6classes,10 orders,19 family,and 24 genera.Up to 87.1%of OTUs can be identified to the level of family and genera,among them 96 OTUs could be identified to the level of family,top 3 families were Sebacinaceae.Thelephoraceae.and Pyronemataceae in terms of frequency.127 OTUs could be identified to the level of genus,top 3 genera were Sebacina.Tomentella,and Amphinema.There were much similar on the community structure of ectomycorrhizal fungi between Halawu and Luanchai ditch which shared 70.1%of coincidence of OTUs sequences by the Venn diagram analysis.Compared with traditional way of studying ectomycorrhizae,454 high throughout sequencing could reveal the more diversity of the community structure of ectomycorrhizal fungi.
