Study On The New Stran Of Hedychium Named 'Xinghuangxin'(Hedychium Coronarium × Hedychium'Fen') In Vitro Culture And The Selection Of Pot Substrate

There are a great many excellent Ginger Plants,only a few flowers enter the cut flower market and ornamental areas.In the study,regard ‘Xinghuangxin'(Hedychium coronarium × Hedychium ‘Fen'),a new hybrid of Hedychium coronarium and Hedychium ‘Fen',as experiment materials,whose heterosis can make up the flower market flower type peculiar,the fragrant flowers kind of insufficiency,the development garden ground is ornamental new vision has the practical significance.The study explore establishing aseptic systems,multiplication,strength and radication on tissue culture.In the meanwhile,filter out the cultivation medium ratio suitable for plant production.The main result are as follows:(1)The pollution rate is only 35.1% by using 75% alcohol 30s+0.1mg/L mercury bactericidal treatment.The induction rate of callus was 94.9% with the initial flower as explants.The medium MS with NAA 0.3mg/L and 6-BA 0.1mg/L was suitable for inducing callus formation.(2)Take ‘Xinghuangxin' aseptic sheet near the base of the stem,using slice culture technique of induction of adventitious buds,the medium MS with TDA 0.1mg/L was suitable for apply thin tissue.Cluster Bud Multiplication the medium MS with NAA 0.1mg/L and 6-BA 6.0mg/L,the multiplication coefficient is about 3.7(3)A certain concentration of activated carbon can simulate darkness around the root,the best medium for seedling cultivation is 1/2MS+NAA 0.5mg/L+6-BA 0.1mg/L+2g/L activated carbon.As for transplanting,dry the vitro plantlets aged 45 d in the shade for 3-4d,remove the medium under circulating water.Irrigating 1/40 MS inorganic nutrient solution while planting in training ground(peat: sand=1:2),with 90%-94% survival after 20 days.(4)The best cultivation medium ratio for imported peat: pond silt: sand=4:3:1(the pot diameter is 50 cm and the height of 30cm).The aim of this experiment was to establish the mating technique of ‘Xinghuangxin' flower in vitro,to select the suitable proportion of potted substrate,to provide technical support for new variety development and commercial seedling production,to provide theoretical basis for scientific research and to keep good parents for breeding.
