Objective The experiment was conducted to investgate the effect of different energy and protein levels on growth performance and expression of the skin related gene of 15 months Dezhou donkeys.The objective of this experiment was to determine the optimum levels energy and protein of Dezhou donkeys and provide reference for the nutritional requirements of Dezhou donkeys.Method A total of 84 Dezhou donkeys of 15 months were randomly divided into 7 treatments,12 replicates of per treatment(6 female 6 male).The diets with 4 levels of energy E1(2.4 MC/kg),PE(2.6 MC/kg),E2(2.8 MC/kg)and E3(3.0 MC/kg);The diets with 4 levels of protein P1(11.93%),P2(13.95%),PE(15.98%)and P3(18.01%).The serum biochemical indices of Dezhou donkeys was detected with automatic biochemical analyzer,the serum antioxidant indices was measured with ELISA kit and the expression levels of COL1A1,EGF and EGF-R m RNA in skin were determined with real-time PCR technique.Results 1?The result showed that the energy level have significant influence on growth performance(P<0.05),with the increase of energy,the male's ADG in the group E3 significantly increased than that of group PE,The pasture ADFI and concentrate F/G in the group E3 significantly reduced than that of in the group P1;The female's ADG in the group E2 significantly increased and than that of in the group E1,The pasture ADFI climbed up and then declined,which in the group PE was greater than that in the group P3,The F/G in the group E3 significantly reduced than that of in group E1,the Donkey hide weight of group PE significantly reduced than other groups(P<0.05).Briefly,the protein levels have no significant influence on ADG,ADFI and F/G of the Dezhou donkeys(P>0.05),but the Donkey hide weight of group PE significantly reduced than other groups(P<0.05).2?Both the energy levels and protein levels had significant influence on serum biochemical indices of the Dezhou donkeys(P<0.05),with the increase of energy,The serum TP,ALB,CHOL and GLU contents of male's in the group E3 significantly increased than that of in group E1 with the increase of energy level;the content of serum TP and ALB in female's Donkeys climbed up and then declined,especially in group E2.The content of CHOL and GLU in the group E3 significantly increased than that in the group E1;with the increase of protein,male's serum TP,ALB and GLU contents rase at first,then decrease,group P2 was greater than P3;the female's serum TP content same as the male's.3?The energy levels and protein levels have both significant influence on serum antioxidant indices(P<0.05),with the increase of energy,the content of male's serum CAT climb up and then decline,group E2 was highest than other groups;the same to the female's.with the increase of protein,male's serum CAT content decreased significantly,group P1 was higher than other groups;female's serum CAT content same as male's,content of serum GSH-Px rase at first,then decrease,group P2 was greater than group P3.4?The energy levels and protein levels have both significant influence on expression of skin related genes(P<0.05),with the increase of energy,the expression of COL1A1?EGF and EGF-R of skin have the same trend that rase at first,then decrease,group PE was higher than other groups;the female's same as male's.with the increase of protein,the expression of COL1A1 of male's skin increased significant,group P3 higher than group P1,the expression of EGF and EGF-R climb up and then decline,group PE higher than P1;the females have the same trend with males.5?The energy levels have moderate correlation with male's ADG,F/G,EGF and EGF-R and high correlation with ADFI,have moderate correlation with female's ADFI and EGF-R and high correlation with ADG and F/G.the protein levels have high correlation with male's COL1A1 and moderate correlation with F/G,EGF and EGF-R,the female's have the same trend with males except F/G.Conclusions 1?The diet energy level is 2.8 Mcal/kg,protein level is 13.95%,Dezhou donkey's ADG,ADFI and F/G is better than other groups,the serum levels of TP,ALB,CHOL,GLU,SOD,CAT and GSH-Px in best.2?The diet energy level is 2.6 Mcal/kg,protein level is 15.98%,Dezhou donkey's hide weight,the expression of COL1A1,EGF and EGF-R higher than other groups.3?The energy levels have moderate correlation with male's ADG,F/G,EGF and EGF-R and high correlation with ADFI,have moderate correlation with female's ADFI and EGF-R and high correlation with ADG and F/G.the protein levels have high correlation with Dezhou donkeys' COL1A1 and moderate correlation with EGF and EGF-R.
