Objective By studying the effects of Gamma-aminobutyric on the the regulation of feed intake related genes in the hypothalamus neurons of Chicken,aimed to explor the function of Gamma-aminobutyric adjusting the feed intake,we reveal the mechanism of action by adding Gamma-aminobutyric to improve the production performance in Chicken diets.Methods The chicken embryos eggs needed in the experiment were provided by the Liaoning Medical University hatching room.Planed to select the younger chicken embryos,we had taken the hypothalamus to cultivate the hypothalamic primary neurons cells.Selection of well developed cells,was respectively in the medium,adding different concentration of GABA as processing factor and raise the temperature of 40.5?,adopt the method of Real-time PCR of NPY,Ag Rp,GABAA-?1,MC4 R and POMC.Meanwhile,120 21-days-old Dagu chickens were divided into 4 groups randomly,3 repetitions in each group and 10 birds per repetition.These four groups were fed the diet contained gamma-aminobutyric acid 0,5,50,500 mg/kg in basal diet which was mixed.The experiment was conducted in a phase of two weeks.Then the genes m RNA expression were tested of NPY,Ag RP,GABAA-?1,MC4 R and POMC in hypothalamus.Results1.The hypothalamus neuron cells cultured with 39?were compared with the matched group,and the expression of NPY,Ag Rp,GABAA-?1,MC4 R and POMC m RNA was extremely increased(P < 0.01),the expression of NPY,GABAA-?1,Ag Rp and MC4 R m RNA was the highest with the concentration in0.2 mg/m L of GABA;of NPY and POMC m RNA was the highest with the concentration in 0.02 mg/m L of GABA.2.The hypothalamus neuron cells cultured with 40.5? were compared with the matched group,and the expression of GABAA-?1 and POMC m RNA was extremely increased(P < 0.01)with the concentration in 0.02 mg/m L of GABA,extremely reduced with the concentration in 0.2 mg/m L of GABA(P<0.01),respectively reduced to lower than the matched group and or close to.Compared with the matched group,the expression of NPY,Ag Rp and GABAA-?1 m RNA was extremely reduced with the concentration in 2 mg/m L of GABA(P<0.01),except the POMC and MC4 R close to the matched group.3.Adding 5 and 50 mg/kg GABA in diets extremely increased ADG and ADFI(P < 0.01),extremely increased the feed weight ratio as well(P < 0.01);adding 500 mg/kg GABA,the ADG was extremely lower than the matched group(P<0.01),ADFI and the feed weight ratio was extremely increased than the matched group(P < 0.01).Adding 50 mg/kg GABA,the expression of GABAA-?1 m RNA was extremely reduced compared with the matched group(P> 0.05);the expression of Ag RP and POMC m RNA was extremely lower compared with the matched group in roosters group(P < 0.01),however no obvious difference in hens group(P>0.05);the expression of NPY m RNA of roosters and hens group were respectively higher and lower than the matched group,but neither of them appeared obvious difference(P > 0.05);the expression of MC4 R m RNA was extremely higher than the matched group(P<0.01).Conclusions1.Added appropriate concentration of GABA in diets will increase the feed intake and promote the weight of chicken,but will make side-effect with overtop concentration.2.There are both regulating effects of hypothalamus m RNA expression of NPY,Ag RP,MC4 R,POMC and GABAA-?1 in normal temperature and high temperature,and there is difference between roosters and hens,between internal and external.3.Adding GABA in diets would increase the feed intake of chicken,the mechanism to regulate the expression of hypothalamus feed intake related genes.
