The hair follicle is a self-renewal small organ,which has the characteristic of cyclical growth.Its development and cyclical growth is a complex physiological and biochemical process that involves multi-gene participation,close contact and mutual restraint.A variety of factors control this process.In this study,we used Q-PCR and Western blotting to analyze the expression of EDA and EDAR mRNA and protein in the fetal skin of cashmere goat in 10 period(45 days,55 days,65 days,75 days,85 days,95 days,105 days,115 days,125 days,135 days).Moreover,we used shRNA to explore the effects of EDA and EDAR on the expression of related genes.The results are as follows:(1)The results of Q-PCR show that EDA and EDAR were expressed in the fetal skin.Their expression was opposite.The expression level of EDA on 45 days was significantly higher than that of other period and was the lowest on 85 days.Then it had a little change.The change of expression level of EDAR was stable on 45-85 days,and it was the highest on 95 days.However,it was the lowest on 115 days and 125 days.Then it increased on 135 days.(2)Western blot shows that the expression level of EDAR protein on 45,55 days was lower than that in other period.It began to rise on 65 days,reached the highest on 95 days.Then it decreased gradually and increased on 135 days.(3)The effects of sh-EDA on the expression of hair follicle development-related genes show that the expression of BMP2,BMP4,?-catenin,TGF-?2 and Notch-1 were increased and the expression of Noggin and Wnt-10b were reduced.And the effects of sh-EDAR on the expression of hair follicle development-related genes show that the expression of Wnt-10b was increased,however,the expression of BMP2,BMP4,Noggin,?-catenin,TGF-?2 and Notch-1 were reduced.
