With all the same condition controlled,the aim of this study is to find out the potential clue explaining the difference between Jinhua pigs and Landrace,by comparing activity of intestinal digestive enzymes and expression level of intestinal transporter gene.Furthermore,intestinal microflora was compared between two breeds in order to interpret the difference.Lastly,comparison on concentration of short-chain fatty acid in bowel intestine between two breeds was try to figure out the reason resulted in the difference.1 Comparison on Digestive Enzyme Activity and Expression of Small Intestinal Transporter Gene between Jinhua pigs and LandranceThe purpose of this study was to investigate differences in the digestive enzyme activities and the expression of intestinal nutrient transporter between Jinhua pigs and Landrace,to explore the potential relevance of the pig breeds with the abilities of digestion and absorption.The results showed that in the pancreatic tissue,the activity of trypsin in Jinhua pigs is higher than that of Landrace(P<0.05)and the activity of other digestive enzymes were not significant.In small intestine,except the jejunal mucous membrane,the trypsin activity of Jinhua pigs was significantly lower than that of the Landrace(P<0.05);chymotrypsin activity of Jinhua pigs was significantly higher than that of Landrace in the duodenum and ileum(P<0.05),but had no significant difference in jejunum;amylase activity of Jinhua pigs was significantly lower than that of Landrace in the hole small intestine except ileum mucosa;the activity of maltase in Jinhua pigs was significantly lower than that of Landrace(P<0.05);others had no significant differences;lipase activity of Jinhua pigs was significantly higher than that of Landrace(P<0.05).Real time PCR results showed that relative expression of various gene in each intestinal segments were not significantly different.The results suggest that the pig breed affects the digestiveenzyme activity in the small intestine.The activity of digestive enzyme is different between Jinhua pigs and Landrace.2 Comparison of Intestinal Microbial Community between Jinhua pigs and LandraceThe objective of this study was to investigate the differences of the microbial community between the breeds of pig.Denaturing gradient gel electrophoresis(DGGE)finger print revealed that DNA samples extracted from caecal and colonic contents of Jinhua pigs were generally grouped in one cluster,samples belong to Landrace were grouped in another cluster as well.In addition,samples extracted from small intestinal contents could not be clearly distinguished.Except the duodenal contens sample,Principal component Analysis(PCA)and Principal Coordinate Analysis(PCoA)of the DGGE profile showed the the samples from the Jinhua pigs and the samples from the Landrace were scattered in two different groups.Specific bands of two pig breeds in colonic DGGE profile were collected for sequencing.The results indicated that bands of Jinhua pigs were similar to Roseburia intestinalis and Eubacterium oniliforme,while bands of Landrace were similar to Lactobacillus johnsonii NCC and Lactobacillus reuteri,respectively.These results suggest that microbial community in intestine potentially associated with pig breeds.Futhermore,Jinhua pigs might have more bacteria producing butyrate,while the lactobacillus played a substantial role in the intestine of Landrace.3 Comparison on Concentration of Short Chain Fatty Acid between Jinhua pigs and LandraceThe aim of this study was to investigate the differences on the concentration of Short Chain Fatty Acid(SCFA)in the large intestine between two pig breeds.Jinhua pigs and Landrace were selected as experimental animal.Ten pigs of each breed were slaughtered and contents of caecal and colon was used to detect the SCFA and pH value.Noticeably,pH value in the large intestine of Jinhua pig was significantly lower than that of Landrace.The concentration of acetic acid,propionic acid and butyric acid in caecel and colonic content of Jinhua pigs was markedly lower than that of Landrace.Results showed that pig breeds affected pH value and concentration of SCFA in the large intestine.Jinhua pigs was in a lower level on concentration of SCFA compared with that of Landrace.
