Fatty liver hemorrhagic syndrome(FLHS)is a nutritional and metabolic disease caused by fat metabolism disorder and characterized by fatty degeneration and bleeding of the liver.It is a common disease in high yield hens and high-energy and low-protein diet is the main cause of fatty liver haemorrhagic syndrome on the producing laying hens.In recent years,it has been found that AMPK and its network are closely related to the animal body fat metabolism.The present experiments were carried out to observe the regulation of AMPK signaling pathway and FLHS induced by high-energy low-protein diet.A total of 200 Hyline brown laying hens were randomly divided into 2 groups with 5 repeats in each group and 20 birds in each repeat,control group feeding basic diets(metabolic energy 2678.99 kcal/kg,crude protein 15.86%);experiment group feeding high-energy-low-protein diet(metabolic energy 3100.00 kcal/kg,crude protein 12.00%)..The whole trial period lasted 100 days,and the samples were randomly collected for relevant experiments on 20 days,40 days,60 days,80 days and 100 days.Blood,liver and hypothalamus were collected for determining the expression levels of fat metabolism and AMPK signaling pathway related genes of AMPK,LKB1,ACC,HMGR,FAS,GPAT,CPT1,PPAR? and HNF4?.Moreover,liver tissues were subjected to histopathological analysis with optical microscope.Results were shown as followings:1.H&E and oil red stained found that obvious liver damage such as histological structure fuzzy,cell swelling,the nucleus was located at the edge of the cells,fatty degeneration,fat vacuole of varying quantities and sizes in cytoblastema were observed under light microscope.2.The content of TG,TC,LDL and NEFA were increased significantly,and the content of HDL was decreased significantly in serum and liver of experiment group compared with control group(P<0.05).3.The mRNA expression of ACC,HMGR,FAS,GPAT,HNF4? and PPAR? were increased significantly,and the mRNA expression of AMPK?1,LKB1 and CPT1 were decreased significantly in liver and hypothalamus of experiment group compared with control group(P<0.05).Conclusion: Fatty liver hemorrhagic syndrome caused by high-energy low-protein diet can lead to fatty lesions in the liver,inhibit the activation of AMPK,promote the synthesis of fatty acids and cholesterol,inhibit the decomposition of fatty acids and cholesterol.Moreover,improve the cholesterol and lipid deposition in the liver of the laying hens.
