In order to investigate the changes of fatty acid metabolism in fatty liver hemorrhagic syndrome(FLHS)and search for biomarkers in blood.A total of 40 Hyline brown laying hens were randomly divided into 2 groups with 2 repeats in each group and 10 birds in each repeat,control group feeding basic diets and disease group feeding high-energy-low-protein diet.The whole trial period lasted 80 days,and the samples were randomly collected for relevant experiments on 40 days and 80 days.Liver were collected for determining the expression of AMPK signaling pathway related protein and liver tissues were subjected to histopathological analysis with optical microscope.Blood were collected for Serum metabolomics.Results were shown as followings:1.The results showed that the subcutaneous abdominal fat increased,the liver margin was blunt,crisp and yellow fat deposited in the experimental group.H&E found that the nucleus was located at the edge of the cells,fat vacuole of varying quantities and sizes in cytoblastema were observed under light microscope.2.The protein expression of P-AMPKα were decreased significantly in liver of experiment group compared with control group(P<0.05,P<0.01),the protein expression of ACC and P-ACC were increased significantly in 40 d disease group(P<0.05,P<0.01).However the protein expression of P-ACC and P-ACC/ACC were decreased significantly in 80 d disaese group(P<0.05).3.Serum metabonomics analysis showed that there were different metabolites in the disaese group at 40 days.The content of 3-hydroxybutyric acid,oleic acid,isoleucine,linoleic acid,alpha-ketoisocaproic acid,mannose,phenylacetic acid,3-Aminoisobutyric acid,lauric acid,piperine,succinate semialdehyde,octadecanol,catechol were decreased significantly and dioctyl phthalate were increased significantly.Serum metabonomics analysis showed that there were different metabolites in the disaese group at 80 days.The content of threonine,citric acid,phosphate,palmitoleic acid,threonic acid,myristic acid,uric acid,sucrose,oxalic acid,maleimide,itaconic acid,tartronic acid,sarcosine,cetadiol,resorcinol,malonic acid,dihydrocarveol,2-butyne-1,4-diol,1-indanol were decreased significantly and serine,glutamic acid,phenyl beta-D-glucopyranoside were increased significantly.In summary,AMPK signaling pathway conduction disorders in the liver regulates fatty acid with FLHS,fatty acid synthesis and fat deposition were resulting increased.Metabonomic analysis revealed that fatty acid metabolism and amino acid metabolism are disordered in the whole body,and energy metabolism is increased and decomposed.3-hydroxybutyric acid is closely related to 3 – hydroxybutyrate acted as an important biomarkers of fatty acid metabolism,which may also be FLHS potential biomarkers due to under the influence of FLHS changes significantly.
