At present,due to the destruction of the ozone layer,the UV-B radiation on the earth's surface is enhancing.Enhanced UV-B radiation plays an important role in the evolution of organismand crop yield in the earth's surface,and it plays an important role in the agricultural and non-agriculture systems.Enhanced UV-B have an affect on plant growth and development and physiological metabolism.As an important food crop,the yield of wheat is affected by UV-B radiation.Therefore,in order to adapt to the external environment,it is very important to study the molecular mechanism of wheat to response to UV-B radiation and resist UV stress.The effect of enhanced UV-B radiation on the growth and development of plants is not only reflected in the morphological structure,physiological and biochemical aspects,but also in the level of proteome.In this study,we found that He-Ne laser can alleviate the damage of UV-B radiation to plants.Studies on the effects of He-Ne laser(5 mW·mm-2)and enhanced UV-B radiation(10.08 kJ·m-2·d-1)on wheat proteome was less.The leaf is the main organization of photosynthesis,which are directly affected by UV-B and our lab also found that UV-B irradiation can cause "root bending" phenomenon,therefore,using proteomics technology to study the differential expression proteinsof wheat root and leaf that treated with He-Ne laser and enhanced UV-B radiation research and analysis,which can be benefit to find some proteins relating to control and defense UV-B radiation,and further study the molecular mechanism of wheat responsing to UV-B radiation and He-Ne laser..In this study,we used wheat of 79 as the experimental material.Enhanced UV-B radiation treated wheat for different days,then determined the physiological indexes,and screened out the number of days of wheat irradiation.In addition,the effects of He-Ne laser and enhanced UV-B radiation on wheat proteome were analyzed by proteomic techniques.Results show:(1)Use enhanced UV-B radiation to treat wheat,with the increase of days,the damage of wheat seedling were more serious,mainly reflected in the increasing of MDA content and POD activity,while CAT activity first decreased and then increased and decreased,the soluble protein content decreased first and then increased and decreased and the content of solublesugar increased first and then decreased,which indicated UV-B irradiation of the short time(4days)on wheat seedling damage was not obvious,and even promoted the growth of wheat;the long time(5 days)UV-B irradiation on wheat seedling seriously damaged to wheat seeding,which cause the wheat could not complete the self regulation repair.(2)Analyze the differences of the total leaf protein in wheat treating with He-Ne laser and enhanced UV-B radiation by two-dimensional electrophoresis,and the 16 differentially expressed protein pots meeting the quantitative differences to be at least 2 times more accurate and statistically significant at a significant level of 90% were found,then the biological functions of these proteins were found to be related to photosynthesis,redox reactions,glycolysis,and genetics by mass spectrometric analysis and database retrieval for these differentially expressed proteins.So we speculated that differentially expressed proteins may be associated with wheat leaves responsing to the He-Ne laser and enhanced UV-B radiation,which can deep understanding the mechanism of wheat leaves to UV-B stress response and He-Ne laser on the repair.(3)Analyze the differences of the total root protein in wheat treating with He-Ne laser and enhanced UV-B radiation by two-dimensional electrophoresis,and the 16 differentially expressed protein pots meeting the quantitative differences to be at least 2 times more accurate and statistically significant at a significant level of 90% were found and idenfied.By bioinformatics analysis,it was found that these proteins were related to energy metabolism,redox reaction,sugar metabolism,photosynthesis,cytoskeleton and heredity,which showed that the expression of these 16 proteins were the root of wheat to response He-Ne laser and enhanced UV-B radiation,and to some extent,it reflected the response mechanism of wheat roots to UV-B stress and the molecular repair mechanism of He-Ne laser.
