| With the development of economy and industry,the heavy metal pollution of soil is becoming more and more serious.And now has it become a global problem of environmental pollution.Among the various techniques of soil remediation,phytoremediation has more advantages,for example,the efficiency of repair is high and the cost is low,and there is no secondary pollution,which can be widely used.The ability of wild plants to repair the soil is limited,and the method of transgenic can increase the accumulation and tolerance of heavy metals in soil.As one of economic crops,cotton is very important.It contains a lot of fiber,and it occupies an important position in the textile industry.In addition,it can also produce cottonseed oil and cottonseed cake.Cotton is very important for our industry and agriculture.The aim of this paper is to obtain the regenerated system of anti-heavy metal transgenic cotton by the agrobacterium-mediated method.The recombinant plasmid ACBP1-PBI121 was transformed into agrobacterium by the method of preparing agrobacterium competent cells,and we obtained the agrobacterium containing the target gene ACBP1.Then the agrobacterium plasmid was extracted by the kit,and then the plasmid was determined by electrophoresis.It is identified that agrobacterium contains the recombinant plasmid.In this paper,we cultivated sterile seedlings and the hypocotyls of the seedlings,and infected them with agrobacterium tumefaciens(OD600 0.3~0.5)for 5min,then co-cultured for three days,and then sterilized.The best medium for callus induction is MSB+2,4-D 0.1mg/L+KT 0.1mg/L+sucrose 3%,and it is conducive to reduce the browning that 10mg/L Vc is added and the ammonium nitrate is removed,potassium nitrate is doubled in the medium.The best concentration of antibiotic is 200mg/L of ampicillin and 60mg/L of kanamycin.The addition of 0.2mg/L IBA is beneficial for the growth of the regenerated seedlings.The large area of anti-heavy metal transgenic cotton can achieve the purpose of managing heavy metals in the soil,which is of great significance and value for the environmental protection.In addition,as an economic crop,cotton can not only manage the environment but also produce economic benefits.Iron-deficiency anemia is a widespread iron-deficiency disease in the world,seriously affecting human health.Therefore,it is of great significance for the study of iron supplements.Compared with the traditional iron supplements,polysaccharide iron has more advantages,such as less side effects,easy to be absorbed,good effect,low cost,and so on.In this paper,we studyed the quality of polysaccharide iron capsules,which was of great significance to obtain qualified and stable iron supplement.Contents of the study included traits,identification,inspection(weight loss,related substances,residual solvents,wetting,loading differences),determination of content and dissolution.By observing the color and traits of polysaccharide iron capsules,it was found that the contents were pellets,tan,odorless,slightly salty.The polysaccharide iron was identified from three aspects by refering to the pharmacopoeia.It was proved that iron and polysaccharides in polysaccharide iron were not free,but existed in the complex.The test results show that the content of water is less than 9.0%,and the relevant substance is the glucose by determinating forced degradation with high-performance liquid phase and refractive refractive,and gas chromatography measured that ethanol residue is not more than 0.5%,with wetting,qualified capacity difference.The amount of iron in each capsule is 150 mg which was determined by indirect iodometric method.The dissolution method was established by spectrophotometry,and studyed the dissolution conditions(medium,method,sampling point,etc.),the results found that the dissolution is more than 98% at 90 min.Through studying the quality of polysaccharide iron capsules,we established and verifiedthe detection method,which is of great significance for obtaining safe and effective iron supplements. |
