Study On The Choice Behavior Of Potosia Brevitasis Lewis To The Plant Volatile | | Posted on:2018-08-15 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Gong | Full Text:PDF | | GTID:2323330533964573 | Subject:Agricultural Extension | | Abstract/Summary: | PDF Full Text Request | | The Potosia brevitasis Lewis belongs to the Family cetoniidae of Coleoptera.It is distributed throughout China,and has a wide host range.Since 2001 Potosia brevitasis Lewis entered in Xinjiang,rising rapidly as one of the main pests in agricultural production.the adults of Potosia brevitasis feed on the crops、fruits.and the Agriculture,forestry fruit on yield and quality were badly affected.study on the plant volatile effect on the feeding behavior of Potosia brevitasis.It’s result can not only reveal the chemical links between host plant and Potosia brevitasis,and can provide new ways to control insect pests and methods.This article is using dynamic headspace sampling to collect the apple、maize and grape plant volatiles,Then using the gas chromatography-mass spectro metry(GC-MS)analyzed the volatile components.Using gas chromatography-ele ctroanteimographic detection(GC-EAD)to identified the active constituents of Potosia brevitasis,at last using the Electroantennography(EAG)respond to study the three of the most common host plant of Potosia brevitasis.The main results were as follows:1.Using the EAG test to the crude extracts of the apple、grape、maize three plant fruit volatiles and found that the greatest EAG responses of grape fruit in female adult beetles which were 1.82±0.09 m V,the EAG responses of apple fruit were 1.11±0.08 m V,the EAG responses of maize fruit were 0.69±0.06 m V.The EAG responses of female adult beetles were significantly higher in the grape fruit volatiles than in the apple and maize fruit volatiles.2.Using the GC-EAD to identified the active constituents of Potosia brevitasis and combine with GC-MS and confirm the chemical compound names and the proportion.and obtained 16 compounds including Hexanal、2-Methybutyl acetate、2-Methyl-1-butano、E-2-Hexenal、Hexyl acetate、Octanal、1-Hexanol、Z-3-Hexenol、Nonanal、E-2-Hexenol、1-Octen-3-ol、Linalool、Isophorone、4-Allylanisole、2-Phenyl-2-propan、Geranyl acetate.Finally use EAG analysis of 16 plant volatiles.3.The EAG test of Potosia brevitasis to the 16 standard samples,The results was indicative of responses that the female and male Potosia brevitasis to 0.1μg/μl were significantly higher in the Isophorone than any other.The responses of female Potosia brevitasis to 1μg/μl of Nonanal were 2.55±0.09 mv and significantly higher than any other.The responses of female Potosia brevitasis to 10μg/μl of Octanal、Nonanal、Isophorone were nearly equal,and the EAG responses of Isophorone was 3.50±0.16 mv.The male and female Potosia brevitasis from 0.1μg/μl to 10μg/μl,the EAG responses increases with the increasing of concentration.Under the same dose,the EAG responses of female and male Potosia brevitasis to the(Z)and(E)isomers of hex-2-en-1-ol and hex-3-en-1-ol showed Z-3-Hexenol> E-2-Hexenol> E-3-Hexenol> Z-2-Hexenol.The EAG analysis of Potosia brevitasis beetles to plant volatiles showed the female responses was higher than in the male.The Antennal Sensilla of the male and female Potosia brevitasis will probably have gender-related quantity and the difference distribution or the difference of the physiological,led to the gender difference exist to volatiles. | | Keywords/Search Tags: | Potosia brevitasis Lewis, plant volatile s, EAG, GC-EAD | PDF Full Text Request | Related items |
