Potosia brevitarsis Lewis,Coleoptera,Scarabaeidae,Cetoniinae,is a major pest of crops such as grapes,apples,corn,and tomatoes.Based on the preliminary test results of the research group,the composition and content of volatiles in sweet sour liquid were determined by GC-MS.The electrophysiological and behaviour responses of the Potosia brevitarsis Lewis to the different volatiles were determined by electro-acoustic potentiometer and Y-type olfactometer,to screen out the volatile substances with electrophysiological activity and attractant activity.The selected volatiles were formulated into formulas and the olfactory behavioral responses were determined again to determine the optimal formulation,and the ultra-microscopic structure of the male and female antennae of the white-spotted golden tortoise was performed using scanning electron microscopy.The selected volatiles were formulated into a formulation using Formula Uniform Design and then the olfactory behavioral response was measured again to determine the optimal formulation.Observation and comparison of the ultra-microscopic structure of the male and female antennae of the Potosia brevitarsis Lewis using scanning electron microscopy.In order to elucidate the structural basis of differences in electrophysiological responses and olfactory behavioral responses between females and males,and providing a theoretical basis and technical approach for the green prevention and control of Potosia brevitarsis Lewis.The results show that:(1)Composition and daily dynamic of volatiles in sweet sour liquidThe composition of sugar: vinegar: water: alcohol = 4: 3: 2: 1 volatiles in sweet sour liquid was determined by GC-MS.A total of 30 volatile substances were found.With the increase of the standing time,the types and contents of volatiles also changed.Alcohols,aldehydes and lipid volatiles were the main components,among which the lipid volatiles were the most abundant and the content was high.(2)The EAG response of Potosia brevitarsis adults to different kinds of volatilesThe 15 volatiles at the three test concentrations(10 ?g/?L,1 ?g/?L,0.1 ?g/?L)were able to cause the antennae electrophysiological reaction of the female and male Potosia brevitarsis Lewis.There was no significant difference in the male EAG reaction between the three concentrations of the same volatile.The volatiles with significant EAG reaction at 2-3 test concentrations were: isovaleric acid,dihydromyrcenol,2,3,5,6-tetramethylpyrazine,cinnamaldehyde,benzyl alcohol,2-phenylethanol,hexanoic acid,isopropanol,myristic acid,2-cyclopentylcyclopentanone;for females,there was a significant difference in the EAG reaction value between the three concentrations of the same volatiles,2-phenylethanol,which showed a tendency to increase with increasing concentration,volatiles with significant EAG reactions at 2-3 test concentrations were: 2,3,5,6-tetramethylpyrazine,cinnamaldehyde,benzyl alcohol,2-phenylethanol,caproic acid,isopropanol,nutmeg Acid,2-cyclopentylcyclopentanone.There are significant differences in the electrophysiological responses of the male and female adults of Potosia brevitarsis Lewis to the same volatiles.Multiple test concentrations in 2,3,5,6-tetramethylpyrazine,cinnamaldehyde,benzyl alcohol,2-phenylethanol,caproic acid,isopropanol,myristic acid,and 2-cyclopentylcyclopentanone,the female EAG reaction value was significantly higher than that of the male.In general,the EAG reaction value of the female was higher than that of the male.(3)The behavior response of Potosia brevitarsis to different kinds of volatiles and formulasBoth the female and male adults of the Potosia brevitarsis have extremely significant evasiveness to cis-3 hexene-alcohol.There are also different volatiles between females and males.The directional volatiles between females and males are also different.Among them,males have a very significant tropism to hexanoic acid,n-hexanol,1-octene-3-ol and n-hexanal,and cinnamaldehyde and 2-phenylethyl alcohol only reach significant levels.The females have a very significant tropism to the six volatiles: 2-cyclopentylcyclopentanone,caproic acid,octanal,1-octene-3-ol,isophorone and n-hexanal.Among the 7 groups of formulas,males have better tropism to formula 1,formula 2,formula 5 and formula 7,while females have better tropism only for formula 2,formula 5 and formula 7.And formula 2 has the best attracting effect on both male and female adults.(4)The ultra-microscopic structure of the antennae by Potosia brevitarsisThe antennae of Potosia brevitarsis has a total of 10 knots.It consists of the scape,the pedicel,the 5 funicles and the 3 lamellaes.The male antennae(2232.24 ± 81.96 ?m)are significantly longer than the female antennae(2034.82 ± 18.09 ?m).A total of five sensors were found on the antennae,they're sensilla trichodea,sensilla chaetica,sensilla basiconca,sensilla coeloconica?and B?hn bristles.There was no difference in the type and distribution of the sensor between the female and the male,but the length of the male sensilla basiconca was significantly higher than that of the female,and the female sensilla coeloconica I was also significantly higher than the male.
