| The main application of rectal examination and ultrasound on Superovulation experiment of cattle and cow receptor tracking detection and screening,and implement the transplantation of bovine receptor after optimization scheme.Combined with the estrus miscarriage after seventh days of diameter,circumference,area,body The product and the levels of E2,LH,P4 three kinds of changes in hormone levels,for the further study to improve the Holstein heifers for recipients and pregnancy rate of Japanese black cattle embryos laid the foundation.The Study were determined in centeral dairy farm of western animal husbandry and dairy farm of Shuangqiang animal husbandry limited liability companyShihezi’s city Xin’an town in Xinjiang.Based on production practice,The two programs of superovulation on conventional FSH and long-termFSH were compared withthe young Holstein cattle as the receptor techniques to improve the pregnancy rate of embryo transfer.The 18 eliminate dairy cows were selected as object of study in centeral dairy farm of western animal husbandry and dairy farm and injected.Program of long-term FSH were executed with single injectionon 9 dairy cows among them,in addition program of short effective FSH were operated by 8 needle reduction injection.Then these cows were examined by the portable ultrasound scanner at period time after estrus,then reproductive organs ofthese cows after slaughter were observed.The superovulation effect is obvious,there are a lot of ovulation,the follicle results show that short acting FSH scheme for a total of 179 follicles,an average of 19.8 points;ovulation is 82,average 9.1;long-acting PVP-FSH superovulation,follicle total 248,an average of 27.5 points;ovulation was 112,with an average of 12.4.There was no significant difference between the two effects(P>0.05).Corpus luteum of dairy cows were scanned with portable ultrasound apparatus.The ultrasound morphological indexes of corpus luteum were measured in seventh day after estrus and Artificial Insemination and collected by caudal vein with blood taking needle.The hormone content of E2,LH,P4 of blood serum were detected by relevant ELISA kits.In order to researchrelation between on diameter,area,perimeter,volume of corpus luteum in seventh days after oestrus cycle and hormone levels in serum,85 cows were randomly selected for B ultrasonic examination andP4,E2,and LH on serum were detected by ELISA method.In order to select appropriate recipients for improving conception rate of embryo transfer,we investigated imaging index of corpus luteum and measured size of diameter,area,circumference,volume,respectively.The results showedthat 16 head of 85 dairy cows were examinedwith corpus luteumin seventh days after estrus,others dairy cows without corpus luteum were regarded as the control group.The dairy cows with corpus luteum were analyzed relationship between diameter,area,perimeter,volume and hormonal levels of serum,involving estrogen,LH and progesterone according to the index of different groups.The results showed that there were not significant difference between more breeding3 times groupand below breeding 3 timesgroup onindex of diameter,area,perimeter,volume and BCS(P>0.05),however,there were significant difference on different days in postpartum(P<0.05).There were significant differenceimaging index between 1 times AI and 2 times AI(P<0.05).There were not significant difference between body condition score and luteal imaging index(P>0.05).There were significantly in differentE2 and between less than or equal to 3cm and beyond 2.6cm groupon different diametersP4(P<0.05),there were not significantly different on luteal index of volume,area and perimeter between differentgroups(P>0.05).In order to obtain more pure Japanese black cows,199 head heifers were treated estrus synchronization by placing CIDR in vagina,the rate of estrus was 96%(191/199),113 Chinese Holstein heifers among them wereselected as receptor of embyro transfer by clinical symptoms observation,rectal palpation and ultrasound examination.we measured the diameter of CLat seventh days after estrus by ultrasound.The embryosof Japanese black cows were transferred to the recipient.According to different CL diameter,the recipientwere divided into 3 groups,respectively,1cm≤CLD≤1.5cm、1.5cm<CLD≤2.5cm and CLD>2.5cm.These recipient of ET were diagnosed in twenty-third days after ETby ultrasonography to determine whether pregnant or not in order to study on the effect of CL diameter on conception rate.The results showed thatdifferent diameter groups of 1cm≤CLD≤1.5cm、1.5cm<CLD≤2.5cm and CLD>2.5cm,the differences of the pregnancy rate were not significant(P>0.05),the pregnancy ratewere 42.86%(6/14),38.46%(25/65)and 44.44%(12/27),.respectively. |
