The adaptability of animals to the plateau environment is regulated by the relevant genes.JNKs(c-Jun N-terminal kinases)are members of the mitogen-activated protein kinase(MAPK)family,and are involved in the regulation of life activities such as stress and tissue development.In order to analysis the adaptive evolution mechanism of Tibetan sheep on plateau environment,this study studied the effects of Tibetan sheep,Gansu high mountain fine wool sheep and Tan sheep,introns and exons 1 and 8 were analyzed by real-time fluorescence quantitative PCR.The relative expression of JNK1 m RNA in sheep,liver,lung and other tissues was detected by real-time fluorescence quantitative PCR.Adaptability evolution of plateau.The main results and conclusions are as follows:1.AA,BB and AB genotypes and C.1-75T> C mutations were detected in exon 1 of JNK1 gene in three sheep populations,The AA genotype and allele A were the dominant genotypes and dominant alleles of Tibetan sheep,which were significantly higher than those of Gansu high mountain fine wool sheep and Tan sheep.In the exon 8 region,EE,FF and EF genotypes were detected,and there were three base insertion sites,insertions of C.993-47 base A,C.993-40 base C Insertion and insertion of C.993-41 base A,and c.993-52 A> G mutations.Among them,allele F is the dominant allele of Tibetan sheep and Gansu hig h mountain fine wool sheep.The results are consistent with the altitude of the three sheep populations and the stress of the external environment(alpine hypoxia).To this end,it is speculated that the JNK1 gene can be used as a candidate gene for the adaptation of the sheep plateau and the stress response in the plateau environment,and to provide the basic data for the study of adaptive evolution of the Tibetan sheep.2.The expression of JNK1 gene in 18-month-old Tibetan sheep tissues was detected by real-time fluorescence quantitative PCR.The results showed that JNK1 gene was expressed in all tissues of sheep,but the expression of JNK1 gene was different.(P <0.05).There was no significant difference in the expression of fat between the brain and the brain(P> 0.05),but there was no significant difference between the two groups(P> 0.05)But were significantly higher than those in the liver,heart,breast and spleen(P <0.05).
