Pelodiscus sinensis is commonly known as chinese soft shell turtle,and belongs to the genus Trionyx(Trionychidae)of Tesmdinata.The breeding of turtle species developed rapidly at the beginning of this century,and the annual output increased from 32,000 tons in 1996 to 342,000 tons in 2016.However,with the rapid development of aquaculture,aquaculture technology did not form a specification.Chinese turtle breeding industry is facing the quality degradation,the lack of seed dilemma.It is imperative to improve the relevance and efficiency of the breeding of Chinese turtle.1.SNPs Detection of GHRL Gene and Its Association with Growth Traits.To study the association between GHRL gene and growth traits of Chinese Soft-shell Turtle(Pelodiscus sinensis),we detected and found 14 SNPs of GHRL gene by DNA sequencing:C289T G501 T T738C G776 T A841G T885 C T2960C A2987 T G3390A A3857 C G4718A T4820 C A4850C and T4979 C.The genotype and gene frequency of these SNPs in 120 Chinese Soft-shell Turtle were assayed using the method of Bio-Mass Spectrometry.The result showed that all the loci were in Hardy-Weinberg equilibrium in the Chinese Soft-shell Turtle population.In locus C289 T :All the growth data of CT and CC genotype group were significantly higher than that of TT genotype group.(P<0.05)In locus S2:body weight,body length,body width and calipash length of AB genotype group were significantly higher than that of AA genotype group.(P<0.05)In locus G3390T:body length and body width of AG genotype group were significantly higher than that of AA genotype group.(P<0.05)In locus G4718A:body length,body width and calipash length of AG genotype group were significantly higher than that of AA genotype group.(P<0.05)These loci can be candidate molecular markers for Chinese Soft-shell Turtle breeding.2.SNPs Detection of GHSR Gene and Its Association with Growth Traits.In order to explore the effect of GHSR gene polymorphism on growth-related traits in Chinese soft-shelled turtle,200 turtles were randomly selected to screen SNP loci on GHSR gene by direct sequencing.The results showed that four single nucleotide polymorphisms G106 T,A136G,A13482 C and A13526 T were detected.All SNP loci were in Hardy-Weinberg equilibrium(P> 0.05).The body weight,body length,body width,body height and skirt width of AC genotype at A13482 C were significantly higher than AA genotype.(P< 0.05).The SNPs obtained from GHRL gene in this experiment may help the molecular-assisted breeding ofChinese soft-shelled turtle.3.SNPs Detection by RAD-seq and Its Association with Growth Traits.In order to screen the growth-related molecular markers at the whole genome level and explore their association with growth.In this study,26 individuals were sequenced using RAD-seq technology to obtain a fragment of about 6M and a 5.88 M clean fragment.Using the software bwa to compare the samples,the software GATK for group SNP detection.The results showed that the number of SNPs was 8623836,the number of mutations was 308,378,and the number of mutations was 421403.Rs133922 and rs260217,-log10(P)? 7,may be closely related to the growth of the Chinese soft-shelled turtle,and need to be further verified in the larger population.
