Lycium ruthenicum Murr.is a kind of perennial wild plant in arid area of Northwest China,which has strong cold resistance,drought resistance and salt tolerance.Lycium ruthenicum Murr.has become a research topic because it not only can be used as medicine and food,but has high medicinal and economic value,which can be used as a good plant resources restoration of ecological environment in the western desert area and saline alkali land.Nowadays,fruits is small and hard to pick,leading destruction of forest resources.For the purpose of comprehensive understanding and utilization of Lycium ruthenicum Murr.this precious plant resource,improving the Lycium ruthenicum Murr.seedlings breeding and construction speed,this paper focus on:1.Under low temperature stratification and room temperature drying treatment,the seedling of fruit under salt stress,drought stress and different burial depths was studied.2.Under low temperature stratification and room temperature drying treatment,the seedling of seed under salt stress,drought stress and different burial depths was studied.3.The growth rhythm of seed seedling and field seedling of plug seedling.4.Seedling raising experiment with full light spraying and softwood cutting.To provide the data basis for a large amount of breeding and promotion.The results showed:(1)Compared with CK group under the simulated drought stress of PEG,the germination rate of seeds and fruits treated by low temperature stratification was increased by 60.00% and 16.66% respectively.The number of days of initial emergence was shortened by CK for 7 days and 9 days.Compared with burial depth was 1 cm,the germination rate of fruits and seeds treated by low temperature stratification were increased by 10.07% and 26.67% than room temperature.Respectively,the number of days of seedling emergence was reduced by about 3 days.It shows that the low temperature stratification can effectively break the dormancy of seeds and fruits of Lycium ruthenicum Murr..(2)In-0.03 MPa and-0.10 MPa PEG solution,there was no significant difference in seed emergence and CK treatment between seed and low temperature stratified fruit(P>0.05),it showed that: Mild drought stress did not affect the germination of seeds and low temperature stratification of fruits,seedling index of fruits without stratification treatment decreased significantly with drought stress aggravated.(3)Under 0.4% and 1.0% solution of salt stress,germination index of seed and fruit has no significant difference with CK(P>0.05),0.4% and 1.0% solution of salt stress become threshold of fruit and seed seedling.When salt stress were 0.8% and 1.6% compound,germination index has no significant difference with CK between seed and fruit(P>0.05),0.8% and 1.6% compound solution become threshold of fruit and seed germination.However,seeds and fruits could resume germination after drought stress and salt stress release.(4)The seed germination rate was highest when burial depth was 1 cm,no difference(P>0.05)was observed between 1 cm and 3 cm two groups.The highest germination rate was found at the burial depth of 3 cm,which was not different from that of 1 cm and 5 cm(P>0.05).Covered soil or sand should not be too thick,1 cm was the most suitable sowing depth of seeds,the optimum sowing depth of fruits was 3 cm.It can be used directly in the seedling of Lycium ruthenicum Murr.,which is a suitable technique for saving time and labor and raising seedling rate.(5)In early spring,after seedling in greenhouse with hole tray for 48 days,and field panting to big field when the height of seedling reached 11.3 cm,the average height of plant could reach 40.1 cm with maximum 81.0 cm when stopped growing.Fruit ratio was 23.0%.Applying tray hole seedling method can achieve the goals of early spring seedling,properly field planting,fruit maturated in current year,and forming orchard in one year.Twig cottage has the advantages of short cycle seedling,enlarged amount of nursery stock breeding and prolonged period of propagation,which can be widely used in production.(6)The average height of annual seedling by field seed breeding of Lycium ruthenicum Murr is 25.9 cm,the average diameter of the ground of it is about 0.28 cm,the dry weight of the overground part was about 1.558 g,and the root / crown ratio varied from 0.17 to 0.56.The ability of adapting new environment is strong,for moisture and nutrition might change their growth characteristics of Lycium ruthenicum Murr.In the desert environment,it not only has a larger amount of growth,but also its cycle of fruiting was shorten.(7)In cuttage nursery,200 mg/L GGR-6 solution soaking branches for 1 hours has the highest rate of survival(46.7%);100 mg/L GGR-6 solution soaking branches for 1 hours has maximum number of roots;cuttings with larger diameter is much easier to survive and have more roots.Shoot cutting has many advantages,such as short breeding period,large amount of seedling breeding and long breeding season which provides practical experience for the breeding of fine individual plants.
